Sugarcane is a monocotyledonous plant and a very important sugar-yielding crop from Gramineae family.

Some intergeneric hybrids (Sugarcane×Erianthus fulvus) were identified by chromosome counting and RAPD (Random Amplified Polymorphism DNA) technique.

采用体细胞染色体计数和RAPD(Random Amplified Polymorphism DNA)分子标记的方法鉴定甘蔗与蔗茅属间杂交种F1的真实性。

Basing on our early stage research work about collection and description of sugarcane germplasm and the aided project of Yunnan Natural Science Fund (2001 C0015Q), the wild species Erianthus fulvus, which has good characters, cold-resistant, drought-resistant, higher Brix and less chromosome number, was utilized for the first time through intergeneric hybridization with sugarcane cultivar. Its F_1 hybrids were appraised with some new techniques and methods for germplasm enhancement.

本研究在课题组考察采集和保育鉴定甘蔗野生种质资源的前期工作基础上，依托云南省自然科学基金资助项目(2001C0015Q)，首次利用具有特异优良性状(抗旱、耐寒、锤度高、染色体数目少)的蔗茅野生种与甘蔗栽培种或品种进行属间远缘杂交，扩大甘蔗遗传多样性；采用新技术、新方法对蔗茅杂种F_1代材料进行早期鉴定评价研究，筛选具有育种价值的新种质，为进一步选育生态适应性广、抗逆性强、丰产高糖的甘蔗新品种奠定物质基础。

This method can use not only in rice grasshopper but also in the insects of Odonata, Lepidoptera, Tetrigoidea.

The universal primers of ITS1 and ITS4 for fungi were used in amplification of genomic rDNA sequences. A target fragment was obtained by ITS-PCR. Then, the fragment is cloned into the vector of pMD18-T before sequencing. The result of sequencing showed that the fragment is 692 bp in length. The results of Blast in Genbank and the analysis of fungal rDNA systemic development show that the relation between this fragment to the fungi of Sporisorium is closest, while the relation between it to the fungi of Ustilago is much far. It suggests that the name of Ustilago scitaminea for sugarcane smut isn't consistent to the result of rDNA systemic development analysis. It still needs more experiments to support this hypothesis.

用真菌rDNA序列分析的通用引物ITS1和ITS4，通过ITS-PCR得到目的片段，克隆在pMD18-T载体上后进行测序，结果显示该片段的长度为692 bp，与Genebank中已报道的真菌序列进行Blast和系统发育树分析，表明所获得的序列与Sporisorium属真菌具有更高的亲缘关系，而与Ustilago属真菌的亲缘关系较远，这与一直沿用的甘蔗黑穗病菌的命名似乎并不吻合，有待进行更多的研究以证实。
