A series of cholesterol-bearing O-carboxylmethyl chitosan conjugates with different degrees of substitution of the cholesterol moieties and cholesterol-bearing chitosan were synthesized by the succinyl linkages and characterized by Fourier transform infrared, Proton Nuclear Magnetic Resonance (~1H NMR) and elemental analysis. These hydrophobized chitosan derivatives were amphiphilic in nature and their self-aggregation behavior in aqueous media was evaluated by the fluorescence probe technique.

课题的主要研究内容如下：采用化学的方法合成了不同分子结构、不同取代度的系列胆甾醇基壳聚糖衍生物—胆甾醇基壳聚糖和胆甾醇基-羧甲基壳聚糖，并应用先进的分析手段电位滴定、红外光谱法(IR、核磁共振波谱法等对原料、中间产物以及产物进行了结构表征，衍生物分子中的胆甾醇基取代度(3.2％～12.5％)采用元素分析的方法进行检测。

It was found that vegetable compound wax prepared from 16.7% vegetable wax, 18.8%, 58.3%, 4.1% and 2.1% exhibited excellent performance having needle penetration of 68 (1/10) mm, drop melting point of 60℃, kinematic viscosity of 8.13 mm2/s and its oil content was 34.81%. Besides, it was convenient to be used and transported since it was solid at ambient temperature, thus, it was an ideal oil phase material for emulsion explosive.

当植物蜡用量为16.7%、减五线馏分油用量为18.8%、减四线蜡下油用量为58.3%、微晶蜡用量为4.1%、石油树脂用量为2.1%时，制得的植物型乳化炸药专用复合蜡针入度68 （1/10）mm、滴熔点60℃、运动粘度8.13mm2/s、含油量34.81%，其性能优异，常温下呈固态，方便使用和运输，是乳化炸药理想的油相材料。

The factors affect on the degree of substitution of product, yield and apparent viscosity, such as the mol rates of EPTAC with -NH2, reaction temperature, reaction time and the content of water in system have been discussed in detail. FTIR spectrometer and UV spectrometer are used to characterize the structure of the derivative and it is proved that substitution reaction mainly occurs on the -N element .The particle size and morphology of chitosan and quantemary ammonium salt of chitosan are analysised by comparing photos of their scanning electron microscope. The degree of substitution of HACC is determined by acid -base conductometric titration. An error in current formula of DS has pointed out and a correct formula has presented.On the base of experience of above experiment, the reaction conditions of HACC were systematically investigated through orthogonal design, and then the best conditions are found. Water-solubility and dependence of pH are studied.

The results of three antigensmethods were the same, work concentration of serum was the same(l :100).Results:The protein content of three antigens is, 901 strain with 1000ug/ml,801 strain with 960ug/ml and 951 strain with 1100ug/ml. 1、抗原的提取：收集新疆不同地区分离得到的利什曼原虫前鞭毛体，经培养，用0.01mol/L pH7.2的磷酸盐缓冲液漂洗，超声粉碎，比色法测定其蛋白质含量，间接ELISA法滴定出抗原的工作稀释度。2、检测：用提取的三株抗原，分别进行间接ELISA法测定黑热病病人血清和疫区健康人血清抗体，并以肺结核病人血清、非疫区健康人血清做对照，三株抗原所用检测方法一致，血清工作稀释度均为1∶100。
