The results showed that the Saccharopolyspora spinosa SP06081 protoplast yield was the highest under these conditions: the collected mycelium from SP06081 grown in Tryptic Soy Broth medium with 0.2% glycin for 48 h was treated by 0.1 mg/mL lysozyme at 28oC for 20 min, then plated on the R2YE medium with sucrose as osmotic stabilizer, the number of regeneration protoplast was up to 108/mL. The protoplast-regenerated strains exhibited changes in morphology and antibiotic production, 29.3% protoplast-regenerated strains was characterized by loose mycelium and abundant broken branches as did their parent. Among them, 58.2% strains presented the trend to positive variation in spinosad yield, with the highest spinosad yield of up to 582.0 mg/L, 85.6% higher than that of their parent.

结果表明：菌体在添加0.2%的甘氨酸的TSB培养基中培养48 h收集， 0.1 mg/mL 溶菌酶， 28oC作用20 min制备原生质体，将原生质体涂布于以蔗糖为渗透压稳定剂的R2YE培养基中，原生质体再生数目最多，达108个/mL以上；原生质体再生菌株在形态和抗生素产量上产生分化， 29.3%的再生菌株形态上保持与亲本菌株一致，具有菌丝松散，断裂分枝多的特点，其中53.2%的再生菌株多杀菌素产量变异向正方向移动，最高产量达到582.0 mg/L，比亲本菌株提高85.6%。

Copper contaminated soil was selected, Brassica juncea and Elsholtzia splendens plants were used, then simulation experiment, hydroponic culture, column leaching experiment and pot experiment were conducted to study (1) effects of organic chelants on the Cu specition in soil solution and soil soild phase;(2)Effect and mechanism of organic chelants on plant growth and Cu uptake by plant;(3) nutrient addition on plant growth and Cu accumulation;(4) limitation of chelant in soil;(5) Desolving on nutrient and heavy metals in soil and their environmental risk by chelant application and (6) effects of degradable organic chelant and microbial materials on plant growth and phytoremediation efficiency of Cu contaminated soil.

In the past, researches focused only on sole function of bacteria such as tricalcium phosphate or ferric phosphate solubilization. In order to efficiently develop superior phosphate solubilizing microorganisms which are useful on various crops, a team includes Professors C.C. Young, C.C. Tang, S.H. Su, H.J. Chen and C.T. Lo in National Chung Hsing University and National Formosa University was established. Microbial consortiums include phosphate solubilizing bacteria, phosphate solubilizing actinomycetes, phosphate solubilizing yeast, and phosphate solubilizing fungi will be designed in the present study and their capabilities to promote crop growth will be demonstrated. The suitable phosphate solubilizing bacterial formula will also be designed. Consideration for the specificity and adaptation of phosphate solubilizing microorganisms to their host crops, the designed biofertilizers will be used as inoculants to increase the yields of various crops meanwhile reduce the overdose usage of chemical fertilizers.

过去研究溶磷菌主要是在各别溶磷之功能上之研发，如溶磷酸钙及溶磷铁，本计画为缩短不同菌种筛选在不同作物之效应评估工作，以组成一研究团队进行不同类型菌种之应用评估，成员涵盖二个大学的研究团队，包括杨秋忠、谭镇中、苏孝华及罗朝村等教授，分别研究细菌、放线菌、酵母菌及木霉真菌，另在菌剂生产配方由陈鸿基教授研究，以提高多元化应用土壤磷素之有效利用，并达到不同作物之应用效应评估，期以探讨开发不同作物专用之微生物肥料之可行性及问题，以减少化学磷肥之施用，降低土壤劣化提高农民所得之目的。
