A data acquisition and data analysis software for the fast fluorscence kinetics fluorometer of our laboratory is redesigned and recompiled using assembly and BASIC language. The data acquisition rate in the fast rising part of the fluorescence kinetics curves is one order faster (100μs/point) than the old one. It can measure fluorescence parameters, such as Fo and Fi more accurately for calculation of the relative content of inactive PSⅡ, etc.

用N80-BASIC语言结合汇编语言重新编写了本室快速叶绿素动力学荧光计的测控程序，使快速荧光上升曲线的采样速度提高了一个数量级（达到100μs／点），可对Fo、Fi等关键荧光参数进行精确测定，为无活性PS Ⅱ中心相对含量的准确测定奠定了基础。

By researching the original analysis method for CaF2 in fluorspar,this paper discovered its shortcomings,determined three main factors for causing CaF2 analysis results to be lower slightly:determination of total Ca contents,determination of CaCO3 as well as calibrating of EDTA standard titration solution.

通过对萤石中 CaF2的原分析方法的研究，找出其中的不足，确定了导致 CaF2分析结果偏低的三个主要因素：Ca的总量的测定、CaCO3的测定以及EDTA标准滴定液的标定，结合国家标准分析方法，对我们正在使用的分析方法进行改进，以解决这些问题。

RESULTS suc2 gene fragment of about 1.5 kb was amplified and cloned. DNA sequencing confirmed that , compared with the published sequence, the cloned suc2 gene had correct sequ ence except for one point mutation, but that did not alter the amino acid sequence o f t he encoded peptide. By transformation of yeast cells with a constructed gene dis ruption vector, four yeast clones were obtained which showed correct auxotrophy. PCR and DNA sequencing indicated that these yeast clones had the expected genot ype.

结果用 PCR从酵母基因组DNA中扩增出大小约1.5 kb的suc2基因片段，序列测定表明除585位有一点突变外，所得suc2基因与文献报道相同，构建的定位突变载体导入酵母细胞后，得到4株营养型符合的突变体，PCR表明这些突变体中均有suc2基因的同源重组，取其中1株的PCR产物进行序列测定证实了同源重组的发生。

After 4 passages in culture, the MSCs were implanted into 96 foveola with cell density of 0.08,0.1,0.2,0.4,0.6,0.8,1,2,4,6,8,10(x104/ml) respectively. Meanwhile, the MSCs were cultured in Dulbecco's modified Eagle's medium culture medium with different concentration of fetal bovine serum, the 3-4,5dimethylthiazol-2-yl-2,5-diphenyl tetrazelium bromide assay were used to determine the proliferation index of MSCs 72 hours later, the cell cycle were determine by cytometer also.

贴壁筛选法分离、纯化MSCs，取4代以后细胞，以不同密度接种于96孔培养板内，分别以不同浓度血清进行培养，72h后，采用四甲基偶氮唑盐比色法测定MSCs增殖能力并用流式细胞仪测定细胞周期。
