Methods compare with sysmex-r3500 analyzer、coulter maxm analyzer、miller ocularto determine 130 ordinary samples.results f=3.87,p.05,the difference is conspicuous.each"r"is 0.7331,0.7231,0.5011,each other are beeline correlation.about sysmex-r3500 analyzer,the inter-assay cv of quality control cv=2.95%,the sample is cv=4.42%;about couter maxm analyzer,the intra-assay cv is 5.70%,intra-assay cv is 17.02%;about miller ocular,the inter-assay cv is 26.15%.conclusion sysmex-r3500 analyzer is good at repeatability,convenience,quickness,so it is adapt to clinical batch samples quickly determine.

分别用sysmex-r3500分析仪，coulter maxm分析仪以及国际血液标准化委员会所推荐的miller窥盘法进行比较，对130份正常人标本进行网织红细胞测定。结果：f=3.87，p.05，差异有显著性。其两两之间r值分别是0.7331，0.7231，0.5011，两两之间直线相关。sysmex-r3500分析仪的质控物批内平均cv=2.95%，标本批内平均cv=4.42%，批间平均cv=3.42%；coulter maxm分析仪批内平均cv=5.70%，批间平均cv=17.02%；miller窥盘法批内平均cv=26.15%。结论：sysmex-r3500分析仪具有重复性好，简便，快捷的特点，尤其适合临床大批标本的快速测定。网织红细胞 miller；窥盘法；仪器法

METHODS: The BEC were preincubated with crocetin (1, 0.1 and 0.01 μmol/L) for 12 h, then exposed to AGE (100 mg/L). The RAGE mRNA expression was detected by RT-PCR analysis, the intercellular adhesion molecule-1(ICAM-1) was measured by ELISA. The extracellular superoxide ion and thiobarbituric acid reactive substances were assessed with superoxide ion kit and colorimetric assay, respectively The intracellular I2O2 was also detected using the probe 2,7-dichlorofluorescein. The mitochondrial membrane potential and mitochondrial Succinate dehydrogenase were analyzed by the retention of rhodamine 123 (Rh123) and MTT.

不同浓度的西红花酸(1、0.1、0.01μmol/L)预孵BEC细胞12h后，用AGE(100mg/L)刺激细胞12h，RT-PCR法测定RAGEmRNA的表达水平；ELISA法测定细胞间黏附分子－(ICAM-1)的表达；试剂盒分别检测胞外超氧阴离子和硫代巴比妥酸反应产物浓度；同时，还用2，7-二氯荧光素测定了胞内H2O2的浓度，并用罗丹明123(Rh123)荧光法及MTT法分别检测细胞线粒体膜电位水平和其琥珀酸脱氢酶的活性。

Research results indicate that continuous Regional Identification Function is used in nodal virtual flux method, volumn integral and surface integral enhances the convergence and stabilization;using 'put a big number' to deal with the first boundary is faster than 'Remove row and column', the more the first boundary nodes are the more obvious advantage will display;PCG method is quite faster than Crout method to solve equations;it is reliable to use three-dimensional finite element simulation theories to simulate and predict deep foundation pit dewatering in such areas.

研究结果表明，在结点虚流量法及计算体积分和面积分时引入连续的区域识别函数增强了解的收敛性和稳定性；处理已知水头边界的&置大数&法在时间上要比&去行去列&法短一些，定水头边界节点越多，其优越性表现越明显；PCG法比直接解法快很多；三维有限元数值模拟理论用于模拟预测此类地区的深基坑降水具有较高的可信度。

Remove row and column' and 'put a big number' have been used for the first boundary;a continuous Regional Identification Function is used on the basis of non-variant nodal virtual flux method for the free surface boundary;Gauss formula is used to make the curved surface integral of the free surface boundary transformed as the difference between volumn integral and other curved surface integral to calculate the integral item on boundaries, which avoiding finding the position of free surface, while the continuous Regional Identification Function is applied to calculate volumn integral and surface integral;Crout and PCG method is used for solusion.

Methods: Human T cell leukemia cell line Jurkat was chosen as a model. The effect of deguelin on the growth of Jurkat cells was studied by 3-(4,5-dimethyl-2-thiazolyl)-2,5 diphenyl-2H-tetrazolium assay. Apoptosis was detected through DNA fragmentation assay, Hoechst 33258 staining assay and Annexin V/PI double-labeled cytometry. The expression levels of nucleophosmin and nucleoporins, including Nup88 and Nup214, were studied by flow cytometry, Western Blot and reverse transcription-polymerase chain reaction.

A modified analysis method based on the determination of ozone—indigo disulphonate spectrophotometer(GB/T 15437-1995) in gas phase has been developed to detect the concentration of ozone in liquid phase. An analytical condition of λmax=612～615 nm,ε=20 000 L/, pH 2 is obtained by the titration of indigo disulphonate. Compared with potassium iodide method,it shows a higher precision and accuracy,CV＜3.0%. The results also prove that blank experiment will bring about 5.0%systematic error.

在气相臭氧浓度的靛蓝二磺酸钠分光光度法(GB/T15437-1995)基础上提出了液相臭氧浓度的测定方法，采用靛蓝二磺酸钠标准曲线法代替采用易分解的臭氧标准曲线法，得出测定条件λmax=612～615 nm、ε=20 000 L/、pH为2，通过碘量法比较实验，说明新的测定方法具有较高的精度和准确度，相对标准偏差小于3.0%；比较结果表明忽略空白实验将引入5.0%的系统误差。
