To establish a MPLC method for the content determination of resveratrol in Polygonum cuspidatum Sieb. Et. Zucc, Analytical column SOURCE 5RPC ST 4.6/150 was used with a mixture of acetonitrile+water (30+70,volume ratio ) as mobile phase ,with a flow rate of 1ml/min and the UV detection wavelength was 306nm, The linear range was 0.6-1.6mg/ml (r=0.9919). The average recovery of prodatin was 103.0%-90.4%(RSD=7.21%). The method is convenient and accurate. Result indicated a reverse phase chromatography media based o n polystyrene/divinyl benzene has excellent resolution for resveratrol in cuspidatum. P rovide theoretical support for process development.
色谱条件采用SOURCE 5RPC ST 4.6/150预装柱,以乙腈/水(体积比30/70)溶液为流动相,流速1ml/min、检测波长为308nm,结果表明,白藜芦醇在0.6~1.6mg/ml的浓度范围内峰面积积分值与浓度的相关系数r=0.9919;所测定的虎杖饮片中白藜芦醇平均含量为0.304%,平均回收率为103.0%~90.4%;实验表明,以聚苯乙烯为基架的反相介质对于虎杖中自藜芦醇具有良好的分辨率,适合其有效成分的检测、分离,对虎杖中白藜芦醇制备的科学研究过渡到工艺开拓有很好的借鉴。
