Using the middle cerebral artery occlusion modelin rats and cerebral ischemia-reperfusion models in gerbils and mice, we investigated the influence of pueraria compound on the brain water content and the infarct size, the cerebral apoplexy exponent, the contents of lactic acidand lipid peroxide, the activities of lactic dehydrogenase, glutathione peroxidaseand Na+-K+-ATPase.

采用大鼠大脑中动脉栓塞模型、沙土鼠全脑缺血再灌模型和小鼠脑缺血再灌3种动物模型，研究复方葛根素对脑缺血再灌注脑含水量和梗塞范围、脑卒中指数和脑组织中乳酸、乳酸脱氢酶、过氧化脂质、谷胱甘肽过氧化物酶、Na+-K+-ATP酶的影响。

Methods Myocardial ischemia was induced by subcutaneous injection of ISO (30 mg/kg for two days, once a day). The experimental animals were randomly divided into the normal group, ISO group, positive control group and three crocetin groups (25, 50, 100 mg/kg). Cardiac indexes were examined. The level of LDH, CK, MDA in serum were measured. The content of MDA, GSH-Px and the ATPase activity heart and mitochondria were assayed by colorimetric analysis. The histopathological change of myocardia was investigated by HE staining.

采用大剂量ISOsc诱导产生大鼠急性心肌缺血模型(1SO30mg／kg，每日一次，连续2d)，实验大鼠随机分为正常组、ISO模型组、阳性对照组和3个西红花酸给药组(25，50，100mg／kg)，测定心脏系数和血清中乳酸脱氢酶、磷酸肌酸激酶和丙二醛含量，观察心肌组织谷肮甘肤过氧化物酶、Na^ ，K^ —ATP酶和Ca^2 ，Mg^2 —ATP酶活性及心脏病理改变，观察西红花酸的保护作用。

METHODS: D-galactose (1 000 mg·k-1·d-1 ) was injected into mice hypodermically for 8 weeks to induce aging animal model, and Vit-E (100 mg·kg-1; 250 mg·kg-1) was administered for 6 weeks by ig at the 3rd week of making model. After Vit-E treatment for 8 weeks, water maze test was used to determine the ability of mice's learning and memory. The activities of glutathione peroxidase and succinate dehydrogenase, the content of nitric oxide and activity of nitric oxide synthase in the brain tissue were detected separately.

小鼠连续皮下注射D-半乳糖（1 000 mg·k-1·d-1）8周制备衰老模型，并于第3周开始给予维生素E（100 mg· kg-1；250 mg· kg-1）处理；8周后采用水迷宫测定小鼠学习记忆能力，并取脑组织测定谷胱甘肽过氧化物酶和琥珀酸脱氢酶活性，测定一氧化氮含量和一氧化氮合酶活性。

In addition, SW480 and HCT116 cells were treated with deoxycholic acid for 15, 30, 45, 60, 90 and 120 minutes. It was found that the deoxycholic acid treated cells showed an elevation of ATP activity by 12.35 %, 20.08 %, 29.69 %, 33.56 %, 45.85 %, 54.32 %, respectively. While the HCT116 treated cells, showed an elevation of ATP activity by 3.69 %, 5.59 %, 8.52 %, 12.26 %, 16.52 %, 19.71%, respectively.From the experiments conducted using the cell lines described above, it was further confirmed that genes CYP7A1 (cytochrome P450, family 7, subfamily A, polypeptide 1), AKR1D1 (aldo-keto reductase family 1, member D1), ALDH1A1 (aldehyde dehydrogenase 1 family, member A1), BAAT (bile acid Coenzyme A: amino acid N-acyltransferase), ABCA1 (ATP-binding cassette, sub-family A, member 1), APOC3 (apolipoprotein C-III), LIPA, LIPE (lipase, hormone-sensitive), PTGER2 (prostaglandin E receptor 2) and PTGS2 (prostaglandin-endoperoxide synthase 2) were up-regulated.

此外，为了证实其脂肪代谢产物胆酸对细胞癌化的影响，亦使用SW480和HCT116细胞株，经由脱氧胆酸在15分钟、30分钟、45分钟、60分钟、90分钟和120分钟处理后，其细胞增生比率分别有12.35%，20.08%，29.69%，33.56%，45.85%和54.32%(p.05)以及3.69%，5.59%，8.52%，12.26%，16.52%和19.71%(p.05)上升情形，并从这些细胞株进一步证实CYP7A1(细胞色素P450-7A1)、AKR1D1(醇醛酮还原酶-1-D1)、ALDH1A1(醛脱氢酶-1-A1)、BAAT(胆酸辅酶A：氨基酸N-醯基转移酶)、ABCA1(三磷酸腺苷结合盒转运体A1)、APOC3、LIPA、LIPE(脂肪酶/激素敏感脂肪酶)、PTGER2(前列腺素E受体2)和PTGS2(前列腺素内过氧化物合成酶2)等基因表现皆有呈现向上调控的情形。

Methods: The model of acute myocardial ischemia in rats was developed by ligaturing the left descending anterior coronary artery. Forty male Wister rats were divided into normal control group, ischemia model group, Xuesetong group and Verapamil group. The effect of Xuesetong on the CPK, LDH, SOD and MDA in serum and myocardium and myocardial infarct range was observed.
