The results showed that 1 there were no significant differences in the rates of cytoplast protrusion and enucleation between oocytes that were incubated in colchicine (0.4 μg/mL) for 0.5 h and oocytes that were incubated in colchicine (0.4 μg/mL) for 1 h, and the rate of cytoplast protusion can be 85.4% while the rate of cytoplast enucleation is 100%. 2 There was no significant difference in oocyte enucleation between oocytes treated with medium containing 0.2 μg/mL colchicine for 0.5 h and oocytes treated with medium containing 0.4 μg/mL colchicine for 0.5 h. 3 A maturation time of 18–23 h did not affect the rates of cytoplast protusion and enucleation by chemically assisted enucleation, whereas the rate of enucleation of oocytes by blind enucleation was found to decrease with a prolonged incubation time. 4 The development rates of reconstructed embryos could not be influenced by these two enucleation methods, increased from oocytes matured for 21–23 h.

The oogenesis of P. esculenta can be divided into the following phases in morphological characteristics of oogonium and oocyte development: the proliferative phase of oogonium (0μm in diameter), the initial growth phase of oocyte (10—20μm in diameter), the grand growth phase-Ⅰof oocyte (20—60μm in diameter), the grand growth phase-II of oocyte (60μm×70μm to 120μm×145μm in size), the mature phase of oocyte, and the declining phase, showing a dynamic changes in oogenesis of P. esculenta .

We detected that EGF mRNA was expressed sflungly lii the oocyte, and is also found hi gmnulosa cells, the cell fium smaller foflicular expressed stronger than fium bigger one. In the corpus hemonbaglcwn corpus luteurn, lean type and pseudocorpus-luteum, EGF rnRNA was detected,, no distinct difference can be seen in them. The EGF mRNA expressed strongly in fimbria end, ampulla and isthmus of oviduct, in the big follicular stage, ovulation stage, pregnancy stage and spurius pregnancy stage, we can not see any distinct change in them, but hi the medium follicuar stage,it is weaker.

Results: The results showed that:(1) The three types of NOS immunoreactivity were not found in rat testis until 30 days after birth;(2) NOS1 immunoreactivity was found in a few spermatocytes in rat testes on postnatal 30 days, and in spermatozoa present at the lumen surface of seminiferous tubules and some Leydig cells on postnatal 60 days;(3) NOS2 immunoreactivity appeared in a few spermatocytes, Sertoli cells and peritubular myoid cells on postnatal 30 days. On postnatal 60 days, NOS2 immunoreactivity appeared in some Leydig cells, peritubular myoid cells, Sertoli cells, very few spermatocytes and the head of immature spermatozoa in some seminiferous tubules;(4) NOS3 immunoreactivity was detected in a few spermatocytes on postnatal 30 days, as well as in the smooth muscle cells of blood vessels on postnatal 30 days and 60 days.

In conclusion, these results suggested that (1) the ageing-associated decline in fertility of female KM mouse was due to a decrease in both quantity and quality of oocytes;(2) the ooplasm from young mice did not correct the meiotic errors of aged mice, and the ooplasm from aged mice did not induce abnormal segregation of meiotic chromosome of young mice, indicating that meiotic anomalies found in the oocytes of aged mice might be related to nucleus or chromosomes and relevant factors;(3) a critical nucleocytoplasmic ratio was essential for normal maturation and segregation of meiotic chromosomes of oocytes and development to 2-cell embryos.

Stage I of both male and female stone flounder appears only once in all its life, and the germ cells are comprised by spermatogonia or oogonia. The gonad from Mar to Aug keeps at stage II. The gonad index of testis at this stage is 0.037%, and the amount of spermatogonia is increased quickly. There is some linear germ plasm in the cytoplasm of spermatogina. In ovary at this stage it is mostly composed by oocyte of phase 2 which the character is the appearance of yolk nucleus, and no zona radiate in membrane. The mean GI of ovary is 1.95%. From Sep to Oct gonad is at stage III which testis is composed by lots of spermatogina and few spermatocytes, and the mean GI of testis at this stage is 0.086%. In ovary the ooctyes at phase 3 are in dominate position, the yolk nucleus disappear. And the GI of this stage is 3.35%. Both testis and ovary are at stage IV in Nov. hi testis the germ cells are in spermiogenesis, and the mean GI is 0.93%. hi ovary the oocytes are mostly at phase 4, which are filled in the cytoplasm with vitellin granule, and the zona radiate in membrane begins to formation. Nucleus moves to one side of the oocyte gradually. The mean GI of ovary at this stage is 9.37%.

Pig oocytes cultured in vitro for some time were inseminated by frozen–thawed ejaculated sperm. At specified times after insemination, sperm penetration, cell cycle progression and mitogen-activated protein kinase phosphorylation were evaluated. It was shown that:(1) oocytes at various maturational stages could be penetrated by sperm;(2) sperm penetration did not affect meiotic cell cycle progression;(3) sperm penetration of germinal vesicle oocytes and maturing oocytes did not alter MAPK phosphorylation; and (4) when premetaphase I and metaphase I oocytes, in which MAPK was activated, were fertilised, no evident MAPK dephosphorylation was detected as in metaphase II oocytes.

Japonicus.Nerve extraction substance of sea star and dithiothreitol has been reported to be effectively to induce oocyte maturation in sea cucumbers. In this study we tested their effects on oocyte maturation induction in sea cucumber A. japonicus. The results are as follows, oocyte of A. japonicus does not get mature spontaneously. Neither NES alone nor NES with follicle suspension can induce oocyte maturation. While DTT can prominently increase the percentages of germinal vesicle break down. When the concentration of DTT is between10-1 mol/L and 10-3 mol/L, the effect of maturation induction is more sufficient. When the concentration is about 10-2 mol/L, the percentage of GVBD can reach up to more than 90%.
分别利用在海参中有诱导作用的海星神经提取液和二硫苏糖醇对从成熟卵巢解剖出的刺参卵母细胞进行诱导成熟试验,结果显示,刺参的卵母细胞在海水中不会发生自发成熟,海星神经提取液以及神经提取液加滤泡悬浮液对卵母细胞没有诱导成熟的作用,而二硫苏糖醇处理可以诱导卵母细胞生发泡破裂,当DTT溶液的浓度处于10-1 mol/L到10-3 mol/L之间时,对刺参卵母细胞的促熟作用较为明显,在10-2 mol/L左右时,卵母细胞的诱导成熟率可以达到90%以上。
