Five pairs of primers were designed for PCR according to five fragments of the putative virulent genes acquired by suppression subtractive hybridization between virulent strain HA9801 and avirulent strain 12 of Streptococcus suis type 2 （ SS2 ）. The five fragments were transcriptional regulator (15# and 154# fragments), amino acid permease (210# fragments), ABC transporter (9# fragments) and surface anchored protein (187# fragments).

One L-isoleucine high-producing strain ISW330 with genetic marker was screened through shake tube primary screening, shake flask compound screening, genetic marker test and single clone isolation and continuous passage cultivation This strain could produce L-isoleucine 9.18g/L after fermentation under the fermentation condition without being optimized.

Results: In cellular soluble protein electrophoresis,specific band about 75 KDa was observed in 593 strain.The differences of protein expression at 85,60,50,40KDa between 593 strain and 18 strain were observed.In extracellular soluble protein electrophoresis,specific bands at 75 KDa and 40 KDa were observed in 593 strain;specific band at 50 KDa was observed in 18 strain.The difference of protein expression amount in about 60KDa between 593 strain and 18 strain was observed.

2.3 弓形虫RH株、ZS2株、CN株扩增产物的HpaⅡ酶切鉴定 GRA1基因的酶谱分析表明，HpaⅡ位点在274bp和444bp处，该酶切GRA1后应该有444bp、274bp和170bp 3个片段。
