In this paper, an efficient and easy biological screening method based on the sensitivity to lovastatin of Neurospora crassa was built to deal with the mutant strain rudely by UV mutation. A high-level strain was screened in the shaking flask culture incorporating with TLC and HPLC. A comparison between this two methods also was originally discussed while identifying dyeing using bromophenol blue as the better treatment of TLC, and offering a kind of simple, practical, fast and economical determination qualitatively and quantitatively.

The results Showed that twenty isolates have the following biological aspects:① Capability of forming capsule,② Growth at 37℃ within 3days,③ Forming brown colonies on Niger seed agar, CACA agar,④ Urea hydrolysis test positive within 96 hours.⑤ No blue colonies were found on CGB agar.⑥ The genus could be identified by ID32 identification kid, but the species could not.⑦ The sequence of the LSUrDNA D1/D2 region analyzed confirmed to be C. neoformans var neoformans.⑧ Serotype A and AD were found in these isolates, Serotype A was predominant(83.33%),⑨ Some biodiversities of isolates existed.

Roots of five species of Calanthe and two species of Phaius from north, midland and south of Taiwan were collected for the isolation of orchid mycorrhizal fungi. Total for 104 isolates of fungi were isolated. They are identified respectively by the traditional method or DNA sequence. Results showed thatTrichoderma and Mucor were most often isolated fungi. The DNA sequence results showed that only Xylaris was most common fungus.
