Seeds were sprouted at 25 ℃.The root-tip with 0.5~1.0 cm length were cut and treated in 4 ℃ refrigerator for 24 hours, fixed for 1~1.5 hours with Carnoy's fixative, differentiated for 10-15 min with hydrochloric acid and Ethanol 95 %(1:1), pressed to section in carmine for staining. Them were observed under microscope, then the samples that differentiated well chosen, covered on frozen station with mountin Canada balsam, for micro-photographing, counting and measuring.
