Methods: Twenty-four Kkay mice were divided into three groups(n=8):diabetes group,methionine-diet group,folic acid and vitamin B12 treatment group. C57Bl/6 mice were used as controls(C57). Four months later,the mice were sacrificed and their serum levels of Hcy,folic acid and vitamin B12 were measured by fluorescence polarization immunoassay. Pathological changes in kidneys were examined with the periodic acid schiff method with periodic acid-silver methenamine staining under electron microscope. Expression of MMP-9 protein or mRNA was detected by RT-PCR with immunohistochemical staining. Results: Different morphological changes of DN were found in Kkay mice.

Methods Immunohistochemistry of streptavidin-perosidase method was applied in the observation of the expression of TIAF-1 in each group, including tissues obtained from the unaffected part of nephrectomized kidneys with renal cell carcinoma (n=6) and cadaveric donor of normal kidneys (n=8) as the control group, tissues with acute rejection (AR, n=16) and those with chronic rejection (CR, n=28), and in the analysis combined with the number of positive cells of CD3, CD20 and CD68 infiltrated in the interstitium.

To explore the functions of SPANXA1 in cancerous phenotypes CL1-5 cells were transfected with a SPANXA1 expressing vector and evaluated by cell proliferation, cell migration, Matrigel invasion and colony formation assays in vitro as well as mouse metastasis assays in vivo. The results indicated that the induction of SPANXA1 could reduce cell invasiveness. In the other hand, immunostaining showed that SPANXA1 was predominately located at the nucleoplasm.
