AIM: To isolate bioactive secondary metabolites from the fruits of Schisandra rebriflora Rehd. et. Wils and study their effects on HIV-1 infectivity. METHODS: Inhibition of syncytia formation, HIV-1 reverse transcriptase and protease, protection of HIV-1 infected cells, level of HIV-1 p24 antigen,blockage of fusion were detected. RESULTS: A lignan compound, named rubrifloralignan A, was first isolated as a natural product with modest inhibition effects on syncytium formation and HIV- 1 replication and could protect HIV-1 infected cells with weak inhibition effects on fusion and RT, but it showed no effects on the replication of HIV-1 chronic H9 and PR. CONCLUSION: Rubrifloralignan A is an efficient anti-HIV-1 product with effect on early stage of HIV-1 replication.

In chapter 4, two kinds of amphiphilic amino acids were synthesized andstable liposomes were formed therefrom After the condensationpolymerization of amino acid in bilayer liposomes, stable polypeptide liposomes were obtained, which had lower phase transition temperatureand higher permeability.In chapter 5, four kinds of glycolipids were synthesized and theiraggregation behavior in water was comparied. When incorporated intophospholipid bilayer membranes, they could increase the phase transitiontemperatures and inhibit the aggregation and fusion of mixedliposomesat lower temperature.In chapter 6 and 7, three kinds of steroidal bolaamphiphiles withdifferent chain lengths were synthesized. Incorporation of steroidalmoiety to the center of lipid bilayer membrane obviously increased themobility of lipid membrane and shifted Tc to lower temperature side incomparasion with cholesterol. The bolaamphiphile which was shorter thanthe hosted lipid bilayer membrane thickness influenced the lipid packingmore obviously.

In the present study, we collected cumulus cells oocyte complex from ovaries of two different strain mice. The cumulusenclosed oocytes were cultured for 6 h in MEM supplemented with growth factor and FSH. The meiotic maturation of these oocytes has progressed to pro-metaphse Ⅰ stage and the condensed chromosomes are visible under DIC microscope, metaphase Ⅰ spindle even can be detected under Polscope. The metaphase Ⅰ spindles of oocytes were exchanged under such microscopes. After electric stimuli, 91. 6% and 91. 6% karyoplasts-cytoplasm pairs were fused respectively. The resulting oocytes were cultured further in MEM and over 80% of oocytes released the first polar body. 79% and 77% of oocytes formed two pronuclei after in vitro fertilization and the embryos were cultured in KSOM supplemented with amino acids. Over 60% of embryos developed to blastocyst stage.
