The study of spatial configuration and characteristics of pulsed-wire velocity probe shows that: the yaw-and pitch-response characteristics have a good behavior due to the asymmetrical arrangement of the prongs. Because the amplitude of sensor wire response signal changes with the flow velocity, the differentiated signal is selected to trigger a voltage comparator. The pulsing frequency is restricted to about 100Hz to allow the wire to cool between pulses. The first velocity probe with big acceptance cone has been developed in China, and the way of measuring the Reynolds normal and shear stress and application have been given in this dissertation.

对脉冲热线空间速度探针的空间结构及特性的分析表明：三根长支架平行且安置于同一平面的探针结构可以获得大的测量角和好的测量性能；由于传感丝的响应信号幅值随流速变化较大，故应采用微分法进行信号处理；对于5μm的传感丝由于散热时间基本不变，约为10ms，故脉冲发射的最高频率为100Hz；本文在国内首次研制成功了一种大测量窗的脉冲热线空间速度探针，并给出了测量雷诺应力的方法和实例。

In this thesis, we did modifications of alkyl groups upon benzyl fluorophosphonate. In order to study the influence of alkyl length and steric effect on activity and selectivity of probes, we synthesized benzyl fluorophosphonate recognition heads with modifications including ethyl, butyl, octyl and cyclohexyl. By way of connecting different kinds of tag like azido group, biotin and rhodamine with recognition heads separately through a higher hydrophilic ethylene glycol derived linker, a series of benzyl fluorophosphonate probes were made up. And they will apply to various biological testing strategies.

在本论文中，我们以苯甲基氟化磷酸酯为基础，於其上进行一系列烷基之修饰，为了探讨烷基长度与空间立体性质对於探针活性及选择性的影响，我们合成了包含乙烷基、丁烷基、辛烷基以及环己烷基修饰的苯甲基氟化磷酸酯辨识端，并藉由具较高亲水性的聚乙二醇衍生连接桥结合三种不同的发报端，分别有叠氮基团、生物素及萤光基团来构成不同的化学探针组合，这一系列苯甲基氟化磷酸酯类化学探针将可被应用於不同的生物测试策略。

In the water samples from Gudao, and No. 1 and 2 wastewater stations of the Shengli Oil Field, the ratios of amount of iron bacterial cells to total microbial cells, detected by the combination of the 4 probes and DAPI staining, were 11.0%, 12.8% and 9.0%, respectively. 结果表明，利用基因探针能快速对胜利油田外排水中的铁细菌进行检测：在孤岛采油厂、胜利采油厂1号和2号综合处理站的外排水中，利用4种探针的组合探针检测到的铁细菌细胞数量分别占样品中微生物总细胞数量的11.0%、 12.8%和9.0%，其中Leptothrix和Sphaerotilus，以及Leptospirillum ferriphilum为胜利油田外排水中的3种优势铁细菌，分别占水样中总微生物细胞数的2.94%±0.52%~3.39%±0.52%和2.24%±0.16%~2.63%±0.49%；而Leptospirillum ferrooxidans和Acidithiobacillus spp。

Methods Amniocenteses were performed in 34 pregnant women of 16～23 gestational weeks with indications for prenatal diagnosis. The amniotic fluid samples were cultured in Chang's medium. The metaphase chromosomes were hybridized insitu with the human centromere probes,α-satellites DNA probes of X, Y,13, 21, 18 chromosomes and all primer chromosome probes. These probes were conjugated by Biotin and Dioxin then. The treated slides were examined and taken photos under the fluoromicroscope.
