MIT's arachne is chosen as our assembler. Some problems about software transplant and data preparation are considered. Based on theoretic analysis and data simulation, we have found the main shortcomings of arachne.

我们选用MIT开发的arachne拼接软件作为序列拼接的平台，详细介绍了软件移植和数据准备方面的问题，通过实际结果、理论分析和数据模拟的方法，发现arachne软件拼接血吸虫基因组时存在的主要问题。

Firstly, the key frames were extracted from the video sequence. Then, the features detected between adjacent key frames were guided and matched by motion displacement, which was calculated by the optical flow field. With the RANSAC robust estimation and cascade of homography matrix, the homography matrix of the adjacent key frame and the nonadjacent key frame was got.
