Methods Fifty F-344 rats were divided into four groups, GM only、PAA+GM、PAA only and saline control. ABR thresholds at different frequencies were measured at different times and hair cell losses were numerated. Two-dimensional diffusion assays in the culture medium were performed to evaluate the effect of PAA on the antimicrobial activity.

选用健康F-344大鼠50只，随机分4组：Ⅰ为GM、Ⅱ为PAA+GM、Ⅲ为PAA、Ⅳ为生理盐水对照组；通过观测4组大鼠不同时期、不同频率听性脑干反应（auditory brainstem respons，ABR）阈值的改变；计数耳蜗毛细胞死亡率，以观察PAA对F-344大鼠GM耳蜗毒性的拮抗作用；用双向扩散血清培养基检测法观察PAA 对 GM 抗菌活性的影响。

Nevertheless, as to the antioxidant activity, HA andβ-PPA could also protect cells against oxidative stress within a relatively short duration time when compared to α-Toc. TS didn't show any effect against oxidative stress in the range of our treated doses. In the remanding experiments, all of HA,β-PPA and TS didn't show any activity in the protection of rat primary hepatocyte cells against H2O2-induced oxidative stress, and in the inhibition of telomerase.

Comparing with chicken egg-white lysozyme which is widely used in clinic, hLYZ has many advantages: it is a natural protein in human body, so it is safe and harmless when used as a drug; its bioactivity to kill bacterium is three times higher than chicken egg-white lysozyme, and its thermal stability is much higher, too; it also has many other specific important functions which are not related with its catalysis, such as anti-virus and anti-tumor and improving immunization.

人溶菌酶与目前临床上应用最多的鸡蛋清溶菌酶相比具有以下优势：人溶菌酶是人体内的蛋白，具有天然的相容性，没有毒性和副作用；人溶菌酶比鸡蛋清溶菌酶的杀菌活性高3倍，而且热稳定性也要远高于鸡蛋清溶菌酶；人溶菌酶还有多种特有的与其自身的催化作用无关的理化作用，如抗病毒和抗肿瘤。

Results were shown as followings:(1) Sodium selenite at 0～2.5 μmol/L significantly increased the antioxidative capacity of L-02 cells without having remarkable impact on SMMC-7721 cells;(2) Sodium selenite at concentrations above significantly increased telomerase activity, hTERT gene expression and telomere length of L-02 cells without significant impact on SMMC-7721 cells;(3) Sodium selenite at higher concentrations (larger than 5 μmol/L) resulted in peroxidation of L-02 cells, while scutellarin significantly counteracted its effect;(4) Selenium-rich amino acids from silkworm pupas in the range of 0.5～2.5 μmol/L Se significantly inhibited SMMC-7721 cell growth, induced apoptosis and cell cycle change, and the generation of reactive oxygen species. In contrast, sodium selenite and selenomethionine only had weak impact on them at the same concentrations;(5) A new selenium-containing protein was found from selenium-rich silkworm pupas, which is worthy to be study further;(6) An expression vector containing ansense RNA of hTERT gene were constructed and used to transfect SMMC-7721 cells. They were observed to inhibit hepatoma cells.

结果如下：（1）0～2.5μmol/L亚硒酸钠显著性增强L-02细胞的抗氧化能力；而对SMMC-7721细胞的作用不显著；（2）该浓度硒显著性提高L-02细胞端粒酶活性、增强hTERT基因表达和延长细胞端粒长度；而对SMMC-7721细胞的作用均不显著；（3）高浓度硒（5μmol/L以上）显著性抑制L-02细胞生长、致细胞过氧化，灯盏花素能拮抗硒所致过氧化、降低硒毒性；（4）0.5～2.5μmol/L富硒蚕蛹氨基酸显著性抑制肝癌细胞SMMC-7721生长、导致细胞凋亡和周期改变、诱导细胞产生活性氧，同浓度亚硒酸钠和硒代蛋氨酸对其抑制不显著；（5）富硒蚕蛹蛋白经分离纯化和鉴定后发现存在一新含硒蛋白，其结构和功能有待研究；（6）通过已有的含hTERT基因质粒，成功构建hTERT反义RNA表达质粒，转染SMMC-7721细胞后对其生长具有抑制作用。

By using Morris Maze task,The results indicated the learning and memory impairment was continuously maintained by MA-induced neurotoxity,PF_(11) significantly antagonized the learning and memory impairment by MA-induced neurotoxicity, Motor incoordination was studied by using the rotating rod method in mice,PF_(11) markedly alleviated the injury induced by MA.using HPLC-ECD,there were markedly decrease the contents of DA,DOPAC,HVA and 5-HIAA,PF_(11) markedly antagonised decrease of DA in MA-induced neurotoxicity.

在Morris水迷宫空间辨别性学习记忆实验中，甲基苯丙胺中毒小鼠找到平台的潜伏期明显延长，PF_(11)沈阳药科大学硕士学位论文中文摘要能显著缩短延长的潜伏期。转棒实验表明，甲基苯丙胺中毒小鼠运动协调性显著降低，而PF；；能桔抗小鼠运动协调性的降低。甲基苯丙胺中毒小鼠大脑内多巴胺及其代谢产物DOPAC、HVA含量明显降低，另外，5一羟色胺的代谢产物5－HIAA明显降低，而PF；；与甲基苯丙胺联合给药小鼠大脑内DA、HVA的含量显著升高。这些结果提示PF；通过影响多巴胺神经系统而桔抗甲基苯丙胺神经毒性的作用。
