ABSTRACT: Objective To analyze expression of differentiation antigen of acute myeloid leukemia and its relationship with clinical characteristics and prognosis.

目的 探讨急性髓性白血病分化抗原表达和临床表现和疗效的关系。

Immunohistochemically, cells in myoid/myofibroblastic areas showed positive staining for α-SMA, MSA and vimentin, but negative for desmin and CD34. Electron microscopic study displayed the presence of microfilament bundles, focal dense bodies and micropinocytic vesicles, consistent with those of myofibroblasts.

免疫组织化学标记显示肌样区域细胞表达α-平滑肌肌动蛋白和肌特异性肌动抗原，不表达CD34；电镜观察证实细胞含有质膜下微丝束、局灶性致密体及微胞饮囊泡样结构，与肌纤维母细胞相一致。

Recombinatino polypeptide or chemically synthesized polypeptide antigen is used to immunize the mammal to obtain polyclonal antibody and the nonconservative amion acid sequence of the antiboby to N-terminal with recognition site qBrn-2 of development regulatory protein qBrn-2. The antibody does not recognize other POU structure domain protein, and recognizes specifically qunique protein in Western imprinting experiment. The qBrn-2 expression mode obtained immunohistochemistry process is identical with the resulted height of in-situ molecular hydridization.

With ultrasonic wave and lysozyme, the recombination N protein is split from bacteria. We dissolve it in guanidine hydrochloride, purify it with Ni〓 affinity chromatography column and renature it in vitro. By testing the antigen of the purified N protein, it shows that its activity is very high.
