Now we have known that SARS-CoV encoded 11 varietal proteins. They have 4 constitutive proteins: spike protein, membrane protein, envelope protein and nucleocapsid protein. Spike protein trimer form spike and exsertion? virus peplos on the surface. Spike protein is archi-surface antigen component of virus, it has acceptor binding and membrane fusion activity. Spike protein has important effect at tissue tropism, cell-fusion and toxicity. Nucleocapsid protein can bind genome RNA to shape neucleocapsid. Membrane protein and envelope protein are necessary constituent to shape virus peplos. Spike protein is I-type membrane glycoprotein, it mediate virus binding with host cell epimembranal acceptor and to fuse host cell membrane.

SARS-CoV属于巢状病毒目、冠状病毒科，为单正链RNA病毒，基因组全长约29.7kb，已知编码11种蛋白，其中包括4种结构蛋白，分别是刺突蛋白、包膜蛋白、小包膜蛋白和核蛋白或核壳体蛋白。S蛋白三聚体形成突起，伸出包膜表面。S蛋白是病毒的主要表面抗原成分，具有受体结合和膜融合活性，在组织嗜性、细胞融合和毒力等方面起重要作用。N蛋白与基因组RNA结合形成核衣壳。M蛋白和E蛋白是包膜形成所必需的成分。

In this report, we prepared antiserum to pV of Ad41 and studied the mechanism of Ad41 fastidiousness.

本研究拟表达纯化Ad41 pV抗原，免疫动物制备抗血清，为研究Ad41难养性机理打下基础。

Results The serum antibody level reached to 0.71±0.08-0.77±0.06 after genetic vaccine pCD-HCV1(100 μg/mouse) were inoculated into the mice(n=12) three or four times while blank vector pCD-SRα1 could not induce the mice(n=8) to generate antibody response in same way.After the antibody levels in mice(n=8) immunized by pCD-HCV1 had ascended to peak value(0.71), there was no trend of descending during the following 18 weeks of detection(0.68±0.06-0.75±0.07). Specific fragment of HCV cDNA identified by polymerase chain reaction from DNA extracted from the muscles of the mice after pCD-HCV1 had beed inoculated three months.

结果 基因疫苗pCD-HCV1(100 μg/只)3-4次接种小鼠后(n=12)，血清抗体水平达0.71±0.08-0.77±0.06，空载体pCD-SRα1免疫鼠血清抗体阴性；免疫鼠(n=8)抗体水平达高峰后(0.71)，持续检测18周未见下降趋势(0.68±0.06-0.75±0.07)；pCD-HCV1免疫小鼠3个月后，用聚合酶链反应仍可从肌肉组织中扩增出特异性HCV cDNA片段；免疫鼠PBMC对HCV合成肽CP9、基因重组抗原C、E1刺激后均出现增殖反应，刺激指数分别为4.07±1.58、3.88±0.70和3.69±1.30，与pCD-SRα1免疫鼠相比，差异有显著性(P.001)。

The different fishes which are employed in the present studies are wild-type salmon, cultured salmon of freshwater and seawater, sea perch and fat greenling. The complete CT gene sequences of salmons are obtained by PCR amplification. The partial CT sequences of sea perch and fat greenling are obtained by in vitro cloning PCR method. Alignment of obtained CT sequences with other fish CT shows that CT appears to be well conserved among the same family. And the relative in taxonomy is far away, the similarity of different fish CTs is low. On the contrary, the closer the relative in taxonomy is, the higher the similarity of different fish CTs is. The sCT is expressed in pGEX-4T-X by recombinant form . We also succeed in the research of sCT expression alone by expression PCR. In addition, sCT antiserum is obtained using GST-sCT as antigen, and the high titer is tested by double immunodiffusion. In the rat bioassay, administration of 50 μg recombinant protein evoked significant hypocalcemia at 1 h after the data are analyzed by t-test.

本文用PCR方法克隆了野生鲑鱼、养殖鲑鱼的降钙素基因，并应用体外克隆PCR的方法首次克隆出鲈鱼、六线鱼降钙素的部分基因序列，通过对克隆的降钙素序列的比较研究，结果显示同一科的鱼降钙素序列保守性较高，同时，根据降钙素的部分氨基酸序列进行了降钙素相似性的比较研究，结果显示在分类学上，分类地位较远的鱼，其降钙素相似性较低，分类地位越接近的鱼，其降钙素相似性越高；我们利用谷胱甘肽S-转移酶(Glutathions S-transferase，GST)融合表达载体pGEX-4T-X对克隆的鲑鱼降钙素基因进行了融合表达研究，应用表达PCR的方法对降钙素基因的独立表达进行了初步的研究探索，并将纯化的融合蛋白作为抗原，获得了高效价的兔抗鲑鱼降钙素免疫血清；生物活性研究表明，大肠杆菌表达的融合蛋白具有显著的降血钙作用。
