Objective To investigate the expression of cytotoxic T lymphocyte-associated antigen 4 (CTLA-4) and intranuclear forkhead/winged helix transcription factor p3 (Foxp3) of splenic regulatory T cells in mice, and compare the effects of different stimuli on the immunological activity of Tregs.

目的 研究体外实验中不同细胞刺激剂对小鼠调节性T细胞表面T淋巴细胞毒性相关抗原-4(CTLA-4)及叉头翼状螺旋转录因子(Foxp3)表达的影响，并对不同细胞刺激剂在Treg抑制活性发挥中的作用进行比较。

Bronchial asthma is the most common chronic diseases in children.Asthma can not be fully explained by imbalance of Thl/Th2.With the research progress of CD4+ CD25+ Treg cell,it has been found that CD4+ CD25+ Treg cell related factors such as forkhead/winged helix transcription factor,heine oxygenase-1,transforming growth factor-β,cytotoxic T lymphocyte-associated antigen-4 are closely linked to asthmatic mechanisms.

支气管哮喘是一种儿童时期最常见的慢性疾病，单纯的Thl/Th2应答失衡并不能完全解释支气管哮喘的发生，伴随着CD4+CD25+调节性T细胞的研究进展，人们发现与CD4+CD25+Treg细胞相关的因素如叉头/翼状螺旋转录因子、血红蛋白加氧酶-1、转化生长因子β、细胞毒性T淋巴细胞相关抗原-4与支气管哮喘均有密切的联系。

Results The serum antibody level reached to 0.71±0.08-0.77±0.06 after genetic vaccine pCD-HCV1(100 μg/mouse) were inoculated into the mice(n=12) three or four times while blank vector pCD-SRα1 could not induce the mice(n=8) to generate antibody response in same way.After the antibody levels in mice(n=8) immunized by pCD-HCV1 had ascended to peak value(0.71), there was no trend of descending during the following 18 weeks of detection(0.68±0.06-0.75±0.07). Specific fragment of HCV cDNA identified by polymerase chain reaction from DNA extracted from the muscles of the mice after pCD-HCV1 had beed inoculated three months.

结果 基因疫苗pCD-HCV1(100 μg/只)3-4次接种小鼠后(n=12)，血清抗体水平达0.71±0.08-0.77±0.06，空载体pCD-SRα1免疫鼠血清抗体阴性；免疫鼠(n=8)抗体水平达高峰后(0.71)，持续检测18周未见下降趋势(0.68±0.06-0.75±0.07)；pCD-HCV1免疫小鼠3个月后，用聚合酶链反应仍可从肌肉组织中扩增出特异性HCV cDNA片段；免疫鼠PBMC对HCV合成肽CP9、基因重组抗原C、E1刺激后均出现增殖反应，刺激指数分别为4.07±1.58、3.88±0.70和3.69±1.30，与pCD-SRα1免疫鼠相比，差异有显著性(P.001)。

The conjugate also induced immunological memory.

结果 所制备的多糖衍生物GAMP-ADH及多糖-蛋白结合物GAMP-ADH-Pep10均具有GAMP抗原特异活性，免疫小鼠后可诱生比多糖单独免疫更高水平的GAMP血清IgG抗体，且接近GAMP-ADH-TT疫苗诱生的抗体水平，并能形成免疫记忆。

Results Both GAMP derivatives and GAMP-protein conjugates showed GAMP antigen specificity. Compared with GAMP alone, the GAMP-protein conjugates induced high serum anti-GAMP IgG antibody level and strong bactericidal activity in vitro. The conjugate also induced immunological memory.

结果 多糖衍生物、多糖－蛋白质结合物均具有GAMP抗原特异性；结合物免疫小鼠可诱生较单独多糖更高水平的血清抗GAMP IgG抗体和更强的体外杀菌活性，并能产生免疫记忆。
