Evaluation of IEM103 in rabbit animal model.The evaluation was based on the control-challenge study (IEM101 as control). Rabbits were laparotomized, injecteded 10〓 cfu directly into proximal small bowel, and 28 days later, challenged with 0.25--2ug CT and 10〓-10〓 cfu 4 wild toxic strains with different biotypes and serotypes. Experimental group developed antitoxic and significant vibriocidal immune responses to vaccination, and could protect no less than 2ug CT and 10〓-10〓 cfu all wild strains challenge.

IEM103的免疫原性评价：经兔肠段结扎免疫〓cfu IEM103，7天后在外周血中可检测到外周淋巴细胞分泌性抗CT IgG，血清中可检测到抗CT抗体、杀弧菌抗体，P／N均≥2，且可持续至28天。28天后用0.25-2ug CT毒素，四种毒株滨-43，吴江-2，395，1119〓cfu菌量攻毒，实验组可保护至少2ug CT攻毒，而对照组可保护1ug以下CT攻毒，两组均可保护〓cfu四种毒株攻毒（以肠积液量≥1.0ml／cm为不保护判断标准）。

BACKGROUND: It has been indicated that neuro-immuno-modulation peptide,α-melanophore-stimulating hormone, can play an effective role in anti-inflammation, antibiosis, defervescence and immunological regulation through inhibiting the release of proinflammatory factor, improving the vigor of lymphocyte and other ways, so that it has a good prospect in the prevention and treatment of systemic inflammatory response syndrome.

objective to study the anti-tumor activity and enhancing immune effect of phellinus linteus.methods the anti-tumor activity of phellinus linteus was estimated by the longevity of eac mice,the immune effect of tumorous mice was tested by the experiment of carbon particle expurgation and the il-2 concentration in serum of tumorous mice was determined by elisa.results phellinus linteus can prolong eac mouse's life and can increase the il-2 concentration in serum of tumorous mouse but can't affect the immune effect of tumorous mouse.conclusion chinese hurb phellinus linteus possess some antitumor power for tumor-loaded mice,but further research is needed.

目的 观察桑黄对肿瘤的抑制作用及对机体免疫功能的影响。方法以小鼠艾氏腹水癌的生命延长率评价桑黄的抗肿瘤作用，以炭末廓清实验观察荷瘤小鼠非特异性免疫功能，以elisa法测定荷瘤小鼠血清il-2浓度。结果桑黄对eac荷瘤小鼠的生存时间有延长作用，对荷瘤小鼠血清il-2浓度有提高作用，对荷瘤小鼠非特异性免疫功能无明显影响。结论中药桑黄对荷瘤小鼠具有一定的抗肿瘤能力，作用的强度和作用机制的深度还有待进一步研究。

BALB/c mice were co-inoculated muscularly by this plasmid and the nucleic acid vaccine plasmid pVAXGE expressing the HIV-1 (Human immunodeficiency virus type I) gag-gp120 protein. The level of serum antibodies after immunization showed that the specific antibodies against HIV-1 appeared at the second week and raised to the peak level at the sixth week for the co-immunization group.

Serum and Ig in serum of the three sturgeons can react with serum of rabbit anti each of the three sturgeons' Ig, but cannot react with serum of rabbit anti Ig of carp Cyprynus carpio. The Ouchterlony results show that Ig of sturgeons are immune homologous in structure and sequence, but have little immune homologous characters compared with Ig of high-classification Osteichthyes such as carp.

结果：Fos样免疫反应神经元主要分布在同侧三叉神经脊束核尾侧亚核、双侧延髓内脏带(孤束核、腹外侧区及二者之间的网状结构)。在抗Fos 和抗酪氨酸羟化酶的双重免疫染色切片上，见到延髓内脏带内许多Fos阳性神经元的胞浆同时呈TH样免疫反应(Fos-TH双重阳性细胞)。

Protein-free polysaccharide antigens were obtained after harvest in cells, extraction, deproteinization, dialysis and other steps. Their immunogenicities were verified by ultraviolet spectrophotometer full-wavelength scanning and Western blotting.2、Preparation and identification of rabbit anti-LP1 antibodies and rabbit anti-LP5 antibody Rabbit anti-LP1 and anti-LP5 antibodies were purified after rabbits were immuned with antigens isolated as described above. The purities of both antibodies were above 80% and the titer of blood serum 1:32 tested by double antibody sandwich assay.3、Development of colloidal gold immunochromatographic assay kit The size of colloidal gold particles in the kit was 25nm. The optimal concentrations for antibodies were 30μg/ml and the sensitized concentrations of NC membrane were 5 mg/ml.

主要研究工作从以下几个方面进行：1、LP1~7、9和10型多糖抗原的制备与鉴定将LP1~7、9和10型菌株分别接种在缓冲活性炭酵母琼脂培养基上，37℃、5%CO2的条件下培养，3~5天后洗下菌苔，经抽提、除蛋白、透析等步骤后得到基本无蛋白的LP多糖抗原，经紫外分光光度仪全波长扫描及Western blotting验证其抗原良好。2、兔抗LP1抗体和兔抗LP5抗体的制备与鉴定分别用LP1、LP5型多糖抗原免疫家兔，采用琼脂糖双向扩散试验检测，两种抗体血清效价均为1：32；饱和硫酸铵法提取抗体，SDS-PAGE检定其抗体纯度均达到80%以上。3、胶体金免疫层析检测试剂的初步研制采用柠檬酸三钠还原法制备约25nm大小的胶体金颗粒；分别制备兔抗LP1抗体、兔抗LP5抗体的金标探针，两种抗体的最适标记量均为30μg/ml；选择适当孔径的微孔滤膜为载体包被两种抗体，NC膜包被浓度均为5mg/ml。
