The function of EM65-2 on the RL95-2 was detected by in vivo and in vitro test. We established the protocol of making the human RL95-2 endometrial carcinoma xenografts in the ovariectomized nude mice at first. Secondly we detect the function of EM65-2 on the subcutaneous enografts with or without benzoate estradiol and estrone. The function was evaluated by the change of tumor volume and the weight of the tumor and the uterine at the end of the test.

VEGF的调控分也分为体内和体外两部分进行，体内部分利用前面实验中获得的不同条件下的裸鼠的皮下移植瘤和小鼠的子宫，通过免疫组化方法检测不同雌激素和抗雌激素的条件下的VEGF的表达情况；体外部分，首先建立RL95-2细胞的无血清培养方法，然后建立无雌激素正常pH、无雌激素低pH的培养液和雌二醇浓度为10nM的正常pH等三种条件培养液，于培养的不同时间分离上清液，通过Elisa法检测上清中VEGF的变化。

Prepare 131I-labbelled anti-CD20 monoclonal antibody using lodogen iodine labelling;(2)Measure the immunity activation of labbelled antibody by means of both cell combine analysis and LDH cytotoxicity detection kit;(3)Evaluate the injury rate of tumor exposed to 131I-chimeric anti-CD20 monoclonal antibody in vitro. The MTT method and the experiment of cell growth control are used;(4)Record the apotosis of Daudi cell using gelose electrophoresis;(5)Learn the inhibition effect of radioactive medication on the marrow. Radiohnmunoimages are taken on various intervals after injection of the labeled antibodies to the nude mice. The distribution of radioactive medication, I31l-labeled antibodies, in the marrow tissue indicates the inhibition effect. Here a B cell non-hodgkin lymphoma model in nude mice is established by us;(6)28-day observation are done in 3 groups of nude mice model.

(1) 用lodogen标记法制备131I-国产人鼠嵌合抗CD20单抗；(2)采用细胞结合分析法和乳酸脱氢酶法细胞杀伤性实验测定131I标记后国产人鼠嵌合抗CD20单抗与靶细胞的免疫活和利结合能力；(3) MTT法和细胞生长抑制实验测定131I-国产人鼠嵌合抗CD20单抗的体外杀瘤活性；(4)用琼脂糖凝胶电泳法研究131I-国产人鼠嵌合抗CD20单抗致Daudi细胞凋亡；(5)建立荷人B细胞淋巴瘤移植瘤裸鼠模型，应用γ计数法检测注射到荷瘤裸鼠体内的131I-国产人鼠嵌合抗CD20单抗的组织分布情况，明确其靶向性；(6)将荷瘤裸鼠分3组进行放射免疫治疗，分别为阴性对照组、131I-国产人鼠嵌合抗CD20单抗组、国产人鼠嵌合抗CD20单抗组。

We used Aeromonas hydrophila which we separated from the diseased Paralichthys olivaceus to made a inactivated whole cell bacterin .We injected this vaccine in the mice and the Paralichthys olivaceus .Both the mice and the Paralichthys olivaceus gained a good protection of 80% and 70%.The fish with dipping immersion gained a good protection of 85%,and the incidence of disease was 6%.We detected the antibody level in the antiserum by agglutination reaction and indirect ELISA which seemed to be immunogenic crucially.We anatomized the mice then we found that all the purtenance of the immunzied mice are healty.All of this proved that the vaccine is effective.

本实验将所分离到的嗜水气单胞菌用终浓度为0.2%~0.5%的甲醛对菌体进行灭活，制备了灭活疫苗，并优化灭活疫苗的灭活条件；将制备的灭活疫苗分别注射小鼠和牙鲆，均取得了较为理想的效果，其中小鼠的免疫保护率达到80%，牙鲆的免疫保护率达到70%；采用渗透压差法大规模浸泡免疫牙鲆也取得了很好的效果，浸泡免疫过的牙鲆发病率仅为6%，攻毒后免疫保护率为85%；通过应用间接ELISA方法检测受免动物的抗血清，并将牙鲆的抗血清注射新西兰白兔，从而获得兔抗鱼血清，更便于检测，结果都表明动物体内产生了较高的抗体水平；剖检攻毒后的动物，发现免疫过的动物内脏健康无异样，表明该疫苗安全有效，从而为防治牙鲆腹水症开辟了一条新途径，也为进一步制备其基因工程疫苗奠定了基础。

The experiments may contribute not only to investigating the mechanism of interaction between ldiotype and anti -idiotype T cells by which immune network are up-regulated, but also to analysing the suppressive effect of anti -idiotypic T cell or antibodies against the antigens on the activated T cells in the allo-responsiveness.

本课题选用小鼠耳后心肌移植模型，用同种抗原特异的活化T淋巴细胞免疫同系小鼠的方法，诱导免疫小鼠同种心肌移植物存活时间的延长，以此探讨免疫网络中独特型-抗独特型相互作用机理，着重分析在同种抗原诱导的免疫应答中，抗独特型T细胞和抗活化T淋巴细胞抗原抗体对同种免疫反应的抑制作用。

LPS免疫后第2周实验豚鼠的免疫血清中，抗LPS IgG抗体滴度均明显增高，第3周达高峰，第5周仍维持在峰水平，野生株和突变株LPS免疫血清中的抗-野生株LPS IgG抗体滴度较各自免疫前分别增高8倍和4倍，抗-突变株LPS IgG抗体滴度则较各自免疫前分别增高6.5倍和15倍；(2)全身免疫后第3周和第5周，野生株LPS免疫血清中检测到较免疫前增高6倍的抗-GM1 IgG抗体，而NANA缺失的突变株LPS免疫血清中一直未检测到抗-GM1 IgG抗体；(3)野生株LPS免疫组中有17.3％的坐骨神经原纤维发生以轴索变性为主（占65％）的免疫性损伤，与突变株LPS免疫组和对照组比较均有显著性差异。

Western blots were performed to investigate the cross-reactivity of various labeled antibodies with serum proteins from other fish species. Results Immunoglobulins of five fish species were purified, including bighead carp, carp, argus snakehead fish, ricefield eel, sea bass. The titers of five rabbit anti-sera obtained up to 1:32 examined by agarose double immunodiffusion. Five kinds of purified rabbit anti-fish immunoglobulins antibodies were labeled with HRP and their activities were up to 1:10000 examined by EUSA. Four kinds of rabbit anti-fish immunoglobulins antibodies labeled with HRP could react with serum proteins from other fish species.

结果 纯化了鳙、鲤、乌鳢、黄鳝、鲈五种鱼血清免疫球蛋白，免疫双扩散法测定兔抗这五种鱼免疫球蛋白的抗血清效价均达到1：32，并对纯化的兔抗鳙、鲤、乌鳢、黄鳝、鲈五种鱼类免疫球蛋白的抗体进行了HRP标记，ELISA测定标记抗体的效价达到1：10000左右，Western blots显示标记抗体与部分其他鱼类免疫球蛋白之间存在不同程度的交叉反应。
