In our study, we found menadione has anticancer potential on human breast adenocarcinoma MDA MB 231 by weather cell cycle arrest, anti proliferation, or apoptosis. Low concentration of menadione (5.8μM) significant inhibited growth curve on breast cancer cells (MDA MB 231) but not breast cells (MCF 10A) by performing MTT and trypan blue cell counting. RT-PCR and western blotting showed menadione increase p21 gene and protein expressions but decrease cyclin E and CDK2 protein expression resulting in cell cycle G0/G1 arrest by flow cytometry.

These effects were more significant when adding wtCREG.;③effect on VSMC proliferation: Cell cycle assay and BrDU stain showed: after added the wtCREG and mCREG protein, the ratio of cell in G0/G1 phase increased to 0.5773 and 0.5572 from 0.5308 respectively in OB2 group, which increased to 0.7369 and 0.7034 respectively from 0.6297 in HITASY group;3Role of M6P/IGF2R in CREG biologic function:①ELISA and co-immunoprecipitation showed the wt/mCREG binding to M6P/IGF2R directly.②antibody blocking test: when the anti-IGF2R was added to medium at the same time with wt/mCREG at different concentration(2μg/mL、4μg/mL、8μg/mL),the effects of CREG protein which depressing proliferation, migration, secretion and promoting differentiation were blocked, which had the positive correlation to the concentration of added anti body. The studies showed two combinant CREG promoted VSMC switch to differentiation phaenotype, at the same time, depress VSMC proliferation, migration and secreting extracellular matrix.

The effect of F 1C on joint swelling and the immune respones of AA rats were evaluated by lymphocytotic reaction, flow cytometry, and ELISA. Results F 1C by ig administration to AA rats decreased the primary joint swelling of AA rats, stronger than TW and weaker than CsA, promoted the decreased splenocyte proliferation induced by Con A, and inhibited the augmented thymocyte proliferation induced by Con A in AA rats. The evaluation in vitro showed the F 1C inhibited the augmented thymocyte proliferation from AA rats and promoted the deficient IgG produced by splenocyte from AA rats.
