Objective: To study osteogenic capability of rabbit bone marrow stromal cells and biological characteristical identification in vitro in order to explore the appropriate methods of osteoblast culture in vitro and to select an idea source of seed cells for bone tissue engineer.

目的：研究兔骨髓基质细胞在体外培养条件下的成骨能力及生物学特性，探讨简便易行的成骨细胞体外培养方法，为组织工程选择理想的种子细胞来源。方法：取新西兰白兔骨髓液经离心后得骨髓单个核细胞，以1× 106／ml的细胞浓度进行培养，经传代培养，选择条件培养液培养(1640完全培养液中加入地塞米松10-8Smol／l，β-甘油磷酸钠10mmol／l，VitC 50μg／ml)作为骨髓基质细胞增殖和成骨的调节因子，通过倒置显微镜、HE染色、扫描电镜、碱磷酸酶(alkaline phosphatase，ALP)测定、Ⅰ型胶原免疫组化染色和四环素荧光钙染色等手段对获得的细胞进行生物学特性研究。

The immunohistochemistry results indicate that TGF-β1 plays an important role in the distraction osteogenesis.We expect that we can utilize the ectogenesis TGF-β1 to accelerate bone formation in the near future.

Effects of Adv-hBMP2 gene modify on the bone formation of tissue-engineered bone substitute. BMSCs of canine transferred by Adv-BMP2 gene expressed ALP in vitro and induced ectopic bone formation in nude mice. BMSCs adhered on the surface of FDB after 4hrs, expanded in 24hrs and proliferated in polylayer in 4d. FDB, combined with BMSCs transferred by Adv-BMP2 gene, induced bone formation in the subcutis of nude mice in 6 weeks.

结果：1.BMP-2基因修饰对组织工程化骨成骨的影响 BMSCs经Adv-BMP2基因转染后ALP染色强阳性、并在裸鼠肌肉形成异位骨组织；转基因的BMSCs与冻干骨复合后24小时即完全贴附生长，4天后复层生长并分泌大量胶原，植入裸鼠背部皮下6周后可见大量成骨，很少吸收，而单纯冻干骨或冻干骨复合BMSCs基本不成骨。2。

Abstract］objective: effects of a single period of mechanical strain on gene expression patterns of two osteogenic transcription factors, ets1 and cbfa1, in rat calvarial sutural osteoblast-like cells were studied in this study to elucidate the molecular mechanism of suture distraction.

摘要］ 目的：探讨大鼠颅骨缝成骨细胞在张应力作用下ets1和cbfa1基因表达的变化，以揭示骨缝牵张成骨的分子机制。

Results:(1) CBC possesses a natural trabecular structure. Its components is similar to the mineral of bone.(2) CBC has great cellur biocompatibility. It can promote cell adhesion, proliferation and secretory while needn t modification.(3) In nude mice, the ectopic osteogenesis of MCCAB was obvious which was better than that of CBC alone and the new bone formation increased in pace with time. In the MCCAB, both intramembranous and cartilaginous osteogeneses were seen but the former dominated.(4) When MCCAB was autotransplanted intramuscularly, the osteogenesis was much better than that of negative control groups.(5) The MCCAB can repair autogenous calvaria defects effectively and the effect was much better than that of CBM alone and blank group. Conclusion:(1) CBC possesses good biocompatibility, osteoconduction and porous structure, can be used as scaffold material for constructing of tissue engineering bone.

结果：(1)CBC具有天然的多孔网状结构，最大程度地保持了骨矿的微结构特征，可以作为骨组织工程的支架材料；(2)CBC细胞亲和性好，不需作修饰即能促进成骨细胞的粘附、增殖和分泌活动；(3)MSO/CBC复合人工骨在裸鼠皮下异位成骨作用明显，兼有膜内成骨和软骨内成骨的特点，成骨效果明显优于单纯CBC植入组；(4)MSO/CBC复合人工骨兔自体肌袋异位成骨作用明显，血管化充分，成骨效果明显优于单纯CBC植入组；(5)MSO/CBC复合人工骨修复兔颅骨临界性缺损效果好，明显优于单纯植入CBC组和空白对第四旱区大学硕士学位论文一照组，可以作为骨移植替代材料用于骨缺损畸形修复。
