目的：探讨强骨宝方糖尿病模型鼠含药血清促进体外培养成骨细胞增殖的最佳时相与量效。

OBJECTIVE: To explore the feasibility of adipose-derived stromal cells in repairing loaded bone defects through observing the repair of rabbit tibial bone defects with adipose-derived stromal cells that was cultured to osteoplast and bovine decalcified bone composite.

目的：通过兔脂肪基质细胞诱导培养为成骨细胞，并与牛脱钙骨复合培养，对兔胫骨骨缺损修复情况的观察，探讨脂肪基质细胞修复骨缺损的可行性。

The iliac bone defect moulds were prepared and the compound scaffolds were implanted. The compound scaffolds were taken out after four and eight weeks separately, bone healing status and appearance of scaffolds were observed by eyes, the osteoplast growth status in compound scaffolds were viewed by HE and chinalizarin dyeing separately, the degradation of the compound scaffolds in vivo were observed by SEM.

制作髂骨骨缺损模型，在骨缺损处植入骨支架复合体，植入后4周、8周取材大体观察材料与周围骨愈合情况，HE染色及茜素红染色观察支架复合体内成骨细胞生长及扫描电镜观察支架体内降解情况。

MAIN OUTCOME MEASURES: Morphological changes in primary cultured and subcultured mouse BMSCs were measured. BMSC surface antigen of the second passage of mice (CD105, CD44, CD25, CD34) were determined using flow cytometry. The modified method was assessed to harvest the purity of stem cells. Activity of mouse BMSCs was identified using adipogenic and osteoplastic differentiation. The strength of fluorescent cells following multiple passage was observed.

主要观察指标：原代及传代小鼠骨髓间充质干细胞形态变化；对第2代小鼠骨髓间充质干细胞表面抗原，CD105，CD44，CD25，CD34进行流式细胞仪检测，评价改良方法获取干细胞纯度；通过成脂成骨分化，鉴定小鼠骨髓间充质干细胞的活性；观察多次传代后荧光细胞强度。

After osteoplastic induction, peripheral blood MSCs had strongly positive reactions for alkaline phosphatase staining, total collagen staining, alizarin red staining.

外周血间充质干细胞成骨诱导后，碱性磷酸酶染色、总胶原染色及茜素红染色均呈强阳性；成脂诱导后油红染色呈阳性，有一定数量的脂滴形成。
