METHODS: Total RNA was isolated from hepatocellular carcinoma cell line HepG2. Subsequently, a 1kb fragment of HCCR2 encoding region was amplified by RTPCR and cloned into Promega TEasy vector. After confirmed by DNA sequencing, the fulllength encoding region of HCCR2 was amplified by PCR and cloned into pIRES2EGFP. The recombinant eukaryotic expression vector was confimed by DNA sequencing.

The generic data matrix and database of adult morphology of Tenthredinoidea were set up. The polarization of each morphologic character was concluded based on the evolutionary morphology, the inner group comparision and fossils analysis. The generic cladogram of Tenthredinoidea was calculated by PC586 computer using Hennig86 program. A new classification system for Tenthredinoidea was proposed based on the analysis of the cladogram. The tribal cladistic biogeography of the superfamily was analyzed using Diva program.

Nine medium compositions were used to study their effect on I40 using Plackett-Burman design. The production of biomass for I40 was affected in order of yeast extract, tryptone, glucose, and production of protease affected in order of feather, tryptone and yeast extract.

Basic operators of color image morphology are established.The result of the experiment demonstrates that the color vector order has good property.The algorithm of color image morphology based on this order can better keep the peripheral information and hue of the image and has the strong point of unnecessity of exchanging between different color spaces.

实验结果表明提出的颜色向量序性能良好，建立在该向量序上的彩色图像形态学算法能够较好的保持图像的边缘信息和色调，并且亮度均匀，与原图像相比对比度均匀增强，重构开算法的实验结果比原文献中方法有了较大的改进；具有不需要在不同的颜色空间之间转换的优点。

In degraded PSM matching classification, errors modification method of degraded PSM is presented and the matching tensor of every canonical scattering centers is given. In Chapter 4, the theory of wideband millimeter-wave radar target identification is introduced. It is indicated that, for HR radar target identification, description of temoral relations among features and pattern recogntion adaptive to variation of target angles are of key importance. A rule-based pattern recogniton method of sequential reasoning is proposed, which uses a series of rules to describe relations of features variation caused by target angles variation and has the advantages of adaption to unrandom variation and false probability control in classification over traditional statistical pattern recognitiop method. Feature extraction is crucial step in target identification. In range profiles identification application, features are extracted by means of range domain pre-processing algorithm, spacial and amplitude visual computation directly from range profiles and transform algorithm based on range profiles. Visual or transformed features are either sufficiently convinced or necessarily convinced and both of them are effective and robust to range profiles identification.

在第四章，首先对宽带毫米波体制背景下目标识别方法的一些特点进行了阐述并指出，在毫米波雷达目标识别中，特征之间动态关系的描述以及能适应目标姿态角变化的模式识别方法的研究乃是要解决的关键问题；进而提出了一种基于规则库的序贯推理模式识别新方法，在这种方法的研究中，主要包括序贯推理规则库的规则排列与特征选用顺序以及规则库的收敛等问题；这种方法克服了传统的统计模式识别方法特征利用效率不高、难以适应特征值的非随机性变化的缺点，把姿态角变化所引起的特征的变化用一系列规则加以表示，其优点是能适应特征值的非随机性动态变化，并能控制分类过程中的差错概率α；特征抽取是目标识别中的关键步骤，在基于目标距离像的特征抽取方法研究中，提出了距离空间域的预处理算法、距离空间域与幅度域的直观特征抽取方法以及基于目标距离像变换分析的特征抽取方法。

Na-K-Exchanging ATPase is a universal cell membrane protein in higher eukaryotic cells which mediates the anti-concentration gradient exchange of intracellular Na+ for extracellular K+. It plays essential metabolic roles such as maintaining sodium and potassium ion gradients across the plasma membrane. The targeted sequences were amplified by PCR to determine the genome structure of the hithertofore unsequenced portion of the alpha 1 subunit of the human Na-KExchanging ATPase gene which encodes 80-130 aa of its extracellular domain using two different templates, human genomic DNA and a human muscle cDNA library. The PCR products were analyzed by restriction endonuclease digestion and then cloned into a plasmid vector for chemiluminescence sequencing and further analysis.

Na-K-Exchanging ATPase是一种普遍存在于高等生物体内的细胞膜蛋白，主要参与介导K+和Na+在细胞内外之间的逆浓度梯度的转运，并维持一定细胞内外的离子梯度，我们采用聚合酶链式反应方法分别以人基因组DNA及cDNA文库为模板对人Na-K-Exchanging ATPaseα1亚单位基因胞外区约80-130位氨基酸编码序列进行扩增，限制性酶切分析扩增产物，并进行荧光测序，对测序结果进行同源性分析及剪接位点的搜索并对得到的核苷酸序列进行进一步的分析，发现人基因组DNA和cDNA经过扩增后分别得到833bp和195bp两种不同大小的片段Fg，Fc。

The experimental results show the validness of the mechanism analyses.2. According to the generation mechanism of the leak acoustic signal, the characteristics of the leak acoustic signal are investigated. The auto correlation technology is adopted to descript the leak signal characteristics due to the ability to analyze the coherence of time series. A new procedure to identify the leak acoustic signal from the disturbed noises is proposed based on the conjunction of correlation and approximate entropy algorithm.3. The principle and application in leak location of the correlation and the adaptive filter techniques are introduced. The problems of the traditional leak location methods are analyzed.4. In order to establish the leak detection signal model, the leak acoustic propagation characteristics are investigated experimentally.

从泄漏声产生机理出发，分析了管道泄漏处湍流拟序结构与空泡间相互作用的机理，认为在该机理作用下，泄漏声信号产生过程具有&不可重复&的特征；由于相关函数具有分析时间序列拟序结构的能力，且近似熵从统计的角度区别时间过程的复杂性，因此，提出将信号相关分析和近似熵理论相结合的方法，提取泄漏声信号&不可重复&的特征，辨识管道泄漏故障发生；在实际检测现场，由于各种管内和管外固定声源噪声(如阀门噪声、工地施工噪声等)常导致泄漏误判和漏点定位错误，重点分析了管内固定噪声源的产生机理和特征，为复杂干扰环境下的泄漏辨识提供理论支持。

The fragement of 261 bp was 86 percent the same as a part of the original sequence of Vulgare HortI gene, and the 105 bp was 96 percent compared with a part of the triticum monococcum phosphatidylserine decarboxylase gene. Both of them included an open reading frame.

The distribution database and the geographic cladograms of the superfamily was set up. The distribution pattern of the superfamily was concluded and the origination centre of the superfamily was also discussed. We deduced that the South Asia is probably the origination centre of the superfamily. Two new index for zoogeographic analysis, index of high taxa distribution and the possibility index of the origination of a high taxa, were proposed.
