Objective To study the changes in bone marrow megalokaryocyte numbers, platelet function and T-lymphocyte subsets in children with idiopathic thrombocytopenic purpura after treatment and normalization of platelet count.

目的 研究特发性血小板减少性紫癜患儿经治疗血小板数量正常后的血小板功能、巨核细胞数量及T淋巴细胞亚群的变化。

This data suggested that the retardation of MK endoreduplication during ontogenesis might be related to intrinsic modification of MK progenitor cells.

There were location and morphous changing of megakaryocytes in all PV、ET and IMF patients;the incidence rate of JAK2V617F in PV、ETand IMF patients were 80%、26.7%、50% respectively;the mean of leucocytes in patients with JAK2V617F was higher than patiens without JAK2V617F, different stages of reticulin had no impact on the leucocytes in PV and ET patients who were dignosed in the first time.

PV、ET、IMF患者骨髓切片中均有巨核细胞的形态以及定位变化，PV， ET、IMF患者中JAK2V617F的发生率分别为80%、26.7%、50%；有突变患者的白细胞数均值高于无突变患者，初诊的PV、ET患者骨髓中不同纤维增生程度患者的白细胞均值无差别。

Methods: CFU-MK and the model of thrombopenia of aplastic anemia were analysed by methylcellulose culture or with sera from patients with aplastic anemia. The number of megakaryocyte colony-forming unit and the content of CD41+ cells in CFU-MK were measured simultaneously. Results: After 14 days of culture, the number of CFU-MK and megakaryocyte cluster-forming unit of the model groups were significantly less than that of the normal control groups.

In this study, We used longer DD primers in combination with a two-step PCR protocol to overcome these two problems in term of modified methods of Martin and Pardee.K562 erythroleukemia cell differentiation induced by HEMIN or PMA for 36 hours was used as erythriod or megakaryocytic differentiation model in vitro.Total RNAs were extracted from induced and uninduced K562 cells and were applied to mRNA differential display analysis.
