实验室实验

In this study, isolate 23 microbes with lipase activity from Wu-Jie scrap heap at Ilan and Taichung Environmental Protection Section. When the microbes incubator in 50oC, there are isolates B61 and M63 that lipase activity are better than others. Brevibacillus borstelensis SH168 isolated from NTU resterant food waste compost is used as control check. Isolates B61, M63 and Brevibacillus borstelensis SH168 determine the lipase activity by the LB with tributyrin. The lipase activity of isolates B61 and M63 are 2~3 fold higher than Brevibacillus borstelensis SH168. So isolates B61 and M63 are more useful in producing lipase. Isolate B61 can grow at pH 5~9, and at temperature 20oC ~ 60oC; isolate M63 can grow at pH 6~10, and at temperature 30oC ~ 50oC. Isolates B61 and M63 were identified by 16S rRNA. Isolate B61 is Bacillus circulans, and isolate M63 is Bacillus species.

本研究，於宜兰五结垃圾场和台中市环保局厨余堆肥样品中共分离出 23 株具脂肪酶活性菌株， 23 株菌株中经过三油酸甘油酯的洋菜培养基於 50℃培养 5 天后，发现有 2 株菌的脂肪酶活性较强，分别为分离株 B61 和 M63；以本实验室先前自台大女九餐厅蔬果厨余堆肥中所筛选出的耐高温脂解菌 Brevibacillus borstelensis SH168作为本实验对照菌株，将分离株 B61、M63 及 Brevibacillus borstelensis SH168 3 株菌株於含有三酪酸甘油酯液态培养基培养并测定其脂肪酶活性，发现分离菌株之活性皆比 Brevibacillus borstelensis SH168 高出 2~3 倍，认为分离株 B61、M63 适於产生脂肪酶的菌株，分离株 B61 在 pH 5~9 皆可生长，生长温度介於 20 oC ~ 60oC；分离株 M63 在 pH 6~10 可生长，生长温度介於 30 oC ~ 50oC； 2 株分离菌株以 16S rRNA定序结果分离株 B61 为 Bacillus circulans，分离株 M63 为 Bacillus 属。

MATERIALS: Twenty-four unpregnant female Sprague-Dawley rats, 6 months old, SPF gradem weighing 250-300 g, were used to establish models of postmenopausal osteoporosis.

设计、时间及地点：随机对照动物实验，于2005-06/2006-10在教育部口腔生物医学工程重点实验室完成。材料：6月龄未孕雌性SD大鼠24只，SPF级，体质量250~300 g，构建绝经后骨质疏松症动物模型。

The basic of laboratory is for measuring and testing pressure. The main equipments of the lab. are I,II grade piston type pressure vacuometer(Difinity:±0.02%,Test range:0.25Mpa-60Mpa),Digital pressure meter,Test system and microwave tube of dynamic pressure sensor, Intelligent digital pressure verifier.The test for micropressure, low pressure, middle pressure and high pressure can be worked out.

本实验室主要进行压力的计量和测试工作，主要设备有一，二等活塞式压力真空计（准确度：± 0.02%，测量范围： 0.25Mpa— 60Mpa）；数字式压力计；动态压力传感器的标定激波管和测试系统；以及智能数字压力校验仪等仪器；可进行微压，低压，中压以及高压测试实验。

The same phenomenon was also observed in laboratory experiment A. austera bleached within two days in 33℃. The bleaching process included losing the number of zooxanthellae and the reduction of the pigments in zooxanthellae.

而实验室中的白化实验也发现：简单轴孔珊瑚在水温33℃时，第二天即开始白化，而白化时珊瑚不仅共生藻数目会减少，其单位共生藻的色素量也会减少。

Therefore, it is important to ask whether both Oct-2 and NF-κB participate the MEK/ERK dependent G-CSF expression. G-CSFp-(-289~+25)-Luc, CSF-box-deleted G-CSFp-(-145~+25)-Luc, or octamer-mutated Oct-mut-G-CSFp-(-289~+25)-Luc reporter plasmids were used to investigate the importance of CSF-box and octamer in response to LPS.

在G-CSF的启动子上有两段已被发现对其活性相当重要的序列：CSF-box及octamer，根据过去研究及本实验室未发表的实验结果，此两段序列分别由NF-κB以及Oct-2所结合；於是我们推测MEK/ERK讯息传递路径的下游可能透过NF-κB及Oct-2来调控G-CSF表现。
