本研究共包括以下四部分内容：1、老年糖耐量异常人群中脂联素的改变及其影响因素；2、脂联素受体mRNA在Wistar大鼠体内的分布；3、高脂饲养以及吡格列酮对大鼠脂肪组织脂联素及其受体表达的影响；4、吡格列酮对不同浓度脂肪酸培养的HepG2细胞脂联素受体mRNA表达的影响。

The results show that after lone- term load swimming training, the CEC was increased, the plasma ET content was increased and serum NO content was desreased; endothelium ultrastructure was damaged; tragacanth makes the CEC was decreased After large load exercise, the plasma ET content was decreased and serum NO was content was insreased; endothelium ultrastructure was improved.

结果表明：①大运动量运动训练后，大鼠CEC增多，血桨ET含量上升，血清NO含量下降；内皮细胞结构受损。②黄芪使大鼠大运动量训练后CEC减少，血浆ET下降，血清NO含量升高；内皮细胞超微结构改善。

Methods Group A was NS group. The forty two rats which was infected the trichina worm through their gastrointestinal tract were divided into seven groups randomly.

动物分组：正常大鼠6只为空白对照组；感染旋毛虫的IBS大鼠42只，随机均分为7组：直肠放置气囊对照组、0.6 ml组、0.8 sml组、1.0 ml组、1.2 ml组、1.4 ml组和1.5 ml组。

MAIN OUTCOME MEASURES: At 15 days after model preparation, one rat was obtained from each group to underdone morphological observation. At 21 and 30 days after model preparation, 8 rats were killed in each group, hematoxylin-eosin staining and immunohistochemical method were used to detect the expression of the proliferating cell nuclear antigen, and to calculate fibroblast proliferation index.

主要观察指标：于造模后15 d各组取1只大鼠行组织形态学观察；造模后21，30 d，每组各处死8只大鼠，取材后，行苏木精-伊红染色，免疫组织化学检测增殖细胞核抗原的表达，并计算成纤维细胞的增殖指数。

The results showed that hyperhomocysteinemia was induced by restraint stress. The highest CBS enzyme activity was seen in rat livers. A decrease in hepatic activities of CBS was found in restraint stress rats. The 29.4%±2.5% reduction in the activity of CBS was accompanied by a 44.1%± 3.4% decrease in its mRNA level. CBS enzyme activity was slightly elevated in the kidney of stressed rats while it was almost undeterminable in the cardiovascular system.

Methods A total of 60 adult female SD rats weighing about 340g were randomly allocated into 3 equal groups: unoperated control group (n=20), sham-operated group (n=20) and ovariectomied group (n=20). Immunohistochemical methods were used to determine ER-αand ER-βin superficial and deep compartments in the uterus and vagina.

The unoperated sides of the treated animals also served as controls. Six normal rats were treated as normal control group. Three different siRNA plasmid solution containing RC2-Ⅰ, MAFbx-Ⅱ, CON (50μl , 0.8μg/μl)was injected and transfected by electroporation as methods mentioned above, respectively. The changes of RC2 and MAFbx mRNA levels and RC2 protein levels after 3 days were determined by real-time quantitative PCR and Western blot, respectively. On postoperative 2, 3 and 4 weeks, the rate of wet muscle weight preservation, mean diameter of muscle fiber and mean cross-section area of muscle fiber and muscle protein content were checked and then compared between group CON and group RC2 or group MAFbx, respectively. The differences between groups were analyzed by one-way ANOVA. Ultrastructural changes of muscle fiber were observed at 2, 3, 4 weeks postoperation.Results GFP plasmid was efficiently deliverd into muscle by electroporation and robust GFP expression in muscle could be observed more than three weeks. Histology shows that injected plasmid DNA diffuses extensively in muscle tissue.

1、健康雌性SD大鼠18只，随机分为电穿孔组和非电穿孔组，每组9只，制作右下肢趾长伸肌失神经支配模型；EP组为将质粒pEGFP-N1溶液50μl(0.8μg／μl)注射入右趾长伸肌后，立即于两侧腱腹交接处给予电穿孔，电穿孔参数为：电场强度为200V／Cm，脉冲100μs，频率1Hz，施加10次脉冲；NEP组仅质粒pEGFP-N1溶液注射；转染后1、2、3周，荧光显微镜下观察趾长伸肌中GFP的表达情况，转染后1周行Western印迹检测趾长伸肌中GFP蛋白的表达情况，检测和优化体内转染效率。2、健康雌性SD大鼠78只，随机分为失神经对照组、RC2基因治疗组(RC2组)，MAFbx基因治疗组，每组24只，制作右下肢趾长伸肌失神经支配模型，余6只为正常组；分别将含CON、RC2、MAFbx基因的siRNA重组质粒注射入趾长伸肌，之后给予电穿孔，方法同上；治疗后3天实时定量PCR和Western印迹检测各组中RC2或MAFbx基因的mRNA和蛋白的表达变化，治疗后2、3、4周检测各组肌湿重维持率、肌细胞直径和肌细胞截面积，肌细胞超微结构变化以及肌纤维中蛋白含量变化。
