METHODS: Adult male Wistar rats, 9 weeks of age, were injected intraperitoneally with STZ to create diabetes. At 2 week after the models were created, BDNF protein was administered into the vitreous cavities of rats. At 4 week after the models were created, the rats were killed and the retina was removed for Western blotting and Whole-mount immunohistochemistry for TH to observe the changes of TH and dopaminergic amacrine cells in retina.

9周龄Wistar大鼠，链脲佐菌素（streptozotocin，STZ）腹腔注射，制成糖尿病模型，于模型建立2wk后，大鼠眼玻璃体内注射BDNF溶液，于模型建立4wk后，处死大鼠，取出眼球，取视网膜组织进行Western blotting及视网膜铺片免疫组织化学染色检测，观察视网膜中TH水平的变化，从而反映糖尿病大鼠视网膜中多巴胺能无长突细胞水平的变化。

For this purpose, genotyping was performed on males of 18 Angus, 23 Jinnan cattle, 20 Limousin, 28 Luxi cattle, 26 Qinchuan cattle, 29 Simmental, 29 Charolais. Results Acute oral LD50 were 278 mg/kg and 56.2 mg/kg in rats and mice respectively, while dermal LD50＞2 000 mg/kg and inhalative LC50＞3 000 mg/m3. Skin and eye irritancy were estimated to be negative and mild respectively. Cumulative coefficient was 2.3 and allergic rate was 0%. A negative result was observed in Ames assay while increased frequencies of bone marrow micronuclei and spermatocyte chromosome aberrations were noted in the high dose (11.24 mg/kg) group.

结果 大小鼠急性LD50分别为278 mg/kg和56.2 mg/kg；大鼠经皮LD50＞2 000 mg/kg；急性吸入LC50＞3 000 mg/m3；无皮肤刺激性，轻度眼刺激性；蓄积系数2.3；致敏率为0；Ames试验结果阴性；微核和染色体在高剂量组(11.24 mg/kg)与阴性对照组相比有高度显著性差异(P＜0.01)；大鼠90天亚慢性经口的最大无作用剂量为2.825 mg/kg。
