Rat glioma cell C6 was infected by the deficiency recombinant retrovirus (named C6TK). The sensitivity of C6TK cells to GCV or ACV increased 450 or 10 times than that of C6 cells respectively. Rat C6 glioma model was successfully built, and SD rats inoculated intracerebrally C6TK cells had normal gliomas. The average survival periods of the rats were about 15 days. However, after treated with ACV, the growth of C6TK tumors obviously regressed, and the survival periods extended to 57.8±8.1 days, especially in rats injected with mixed PA317TK and C6 cells or in situ PA317TK cells, which the tumors nearly disappeared after ACV administration and the survival periods extended to over 120 or 71.4±36.1 days respectively, P<0.001.

The new GV preparation method we developed allowed the best visualization of chromatin distribution after Hoechst or CMA3 staining. Our results showed that GV chromatin configurations are different among different species, and thus, while chromatin condensed into a ring around the nucleolus in porcine and bovine oocytes, no such a ring was observed in caprine oocytes; Porcine oocytes were synchronized at GV1, goat oocytes at GV3n and bovine oocytes at GVf configurations before GVBD occurred, indicating that these configurations are progressive towards final maturation rather than atresia; Changes of chromatin configurations were associated with gene activity of transcription; GV chromatin configurations were affected by the environmental changes such as serum starvation and elevated temperature.

METHODS: Female BALB/c mice were treated with CCK-8 and keyhole limpet haemocyanin, splenocytes were acquired and incubated in vitro with KLH restimulation, the productions of Th1 cytokines, interferon-γ, interleukin-2 and Th2 cytokines, IL-4, IL-5 in cell culture supernatants were detected by enzyme-linked immunosorbnent assay. The mRNA expressions of IFN-γ, IL-2, IL-4 and IL-5 in splenocytes were detected by reverse transcription-polymerase chain reaction. The levels of Th1 antibody IgG2a and Th2 antibodies IgG1 in serum were also detected by ELISA.

Methods At first, retrovirus vectors encoding IFN-γ or IL-4 gene were constructed. They were transfected into PA317 packaging cells by lipofectamine, PA317IFN-γ and PA317IL-4 cells were obtained. C6 glioma cells were infected with replication deficiency retrovius containing IFN-γ or IL-4 gene, cell morphology、cell growth curve and cloning efficiency assay were examined. The intracranial C6 glioma animal model was established in immunocompetent Wistar rats. PA317IFN-γ and PA317IL-4 cells were stereotactically implanted into the tumor areas alone or together. The survival of tumor bearing rats were examined, tumor volumes were measured and immunohistochemical analyses of CD4〓 and CD8〓 T lymphocyte infiltration were performed.

Cerevisiae have found that bZIP proteins play a role in yeast anti-oxidation. at3g51960, the transcription factor found in plants, is highly homologous to that of yeast. To examine the in vivo role of at3g51960, the overexpression and silencing vectors were constructed respectively. By using flower dip-dye, the binary expression vectors, pCAMBTA3301: at3g51960 and pCAMBTA3301: at3g51960Promoter, as well as vectors pCAMBTA3301: at3g51960-GFP and pCAMBTA3301: at3g51960cDNA were transformed to Arabidopsis by Tumefaciens. The transgenetic homozygous Arabidopsis strains have been derived after screening. Analysis on these strains reveals that strains overexpressing at3g51960 gene have higher level of stress resistance, implying a role of at3g51960 in plant stress regulation.
