Results: GINaTK retroviral vector containing HSV-tk was constructed and transduced into PA317 retrovirus packaging cell by lipofectin (named PA317TK). Rat glioma cell C6 was infected by the deficiency recombinant retrovirus (named C6TK). The sensitivity of C6TK cells to GCV or ACV increased 450 or 10 times than that of C6 cells respectively. Rat C6 glioma model was successfully built, and SD rats inoculated intracerebrally C6TK cells had normal gliomas. The average survival periods of the rats were about 15 days. However, after treated with ACV, the growth of C6TK tumors obviously regressed, and the survival periods extended to 57.8±8.1 days, especially in rats injected with mixed PA317TK and C6 cells or in situ PA317TK cells, which the tumors nearly disappeared after ACV administration and the survival periods extended to over 120 or 71.4±36.1 days respectively, P ＜0.001.

结果：构建了带有HSV-tk基因的反转录病毒载体GINaTK，应用脂质体转移方法将GINaTK导入反转录病毒包装细胞PA317，成为产病毒细胞PA317TK，用带有HSV-tk基因的复制缺陷型反转录病毒感染C6细胞，命名为C6TK细胞，对GCV和ACV的敏感性分别高于亲本450倍和10倍；成功地建立了大鼠脑胶质瘤模型，并证实转染细胞C6TK的成瘤效应未改变，存活期约为15天；而经ACV治疗后，含有C6TK细胞的肿瘤生长明显被抑制，大鼠生存期延长为57.8±8.07天，尤其是采用PA317TK细胞混和治疗组和原位治疗组，肿瘤基本消失，大鼠生存期显著延长，混和治疗组存活120天以上，原位治疗组存活至71.4±36.1天。t检验，P值均小于0.001。

结果表明：3种处理下，再力花、海寿花和轮叶石胡荽的植株地上部、地下部生物量均以地栽处理最高，植株根冠比均以地栽处理最低；溪荪在3种处理下的适应性没有表现出明显差异；4种植物在3个处理下的根系活跃吸收面积率变化趋势相似，基本在40%～50%波动，没有明显差异；4种植物在泥炭栽培和水苔藓栽培基质下都有一定的适应性。

Results 73 differentially expressed protein peaks were found between SLE patients and normal controls (P＜0.05), in which, 44 peaks had very significant difference (P＜0.01). Detection model of protein mass spectrometry (10542 Da m/z and 2554 Da m/z) was set up by ZUCIPDAS software with the discriminate rate and accuracy rate as 100%, which could distinguish the sample from SLE and normal persons. 10542 Da protein expression peak missed in SLE might be the mixed protein fragments peak of HLA gene family, while the 2554 Da protein expression peak might be a SLE-specific unknown protein.

结果SLE患者治疗前与正常人比较，找出差异表达蛋白峰73个（P.05），其中44个峰有非常显著性差异（P.01），用ZUCIPDAS软件建立的蛋白质质谱检测模型（10542 Da m/z和2554 Da m/z）的判别率和正确率均为100%，可以通过以上两个峰将SLE与正常人区别开来，10542 Da SLE缺失表达的质荷峰可能是HLA基因家族相关的多个蛋白片段的混合峰，而2554 Da可能是SLE特异表达的未知蛋白峰。

The RT-PCR product was inserted into pTG19-T vector and transformed into E. coli successfully. By blastn, the sequence results of Kunming mus musculus were in complete accordance with the conservative sequence of Genbank NR_003278 (791bp-1153bp). By Blastn in NCBI, the sequence with little difference among animals was confirmed to be conservative. After Blastn, fourteen complete CDS coding for different animals were chosen. According to VECTOR NIT 9.0 software, the similarities between Kunming mus musculus and bos taurus, homo sapiens, erinaceus europaeus, cricetulus griseus, sus scrofa, dasypus novemcinctus, rattus norvegicus, rabbit, equus caballus, macaca fascicularis, didelphis virginiana, monodelphis domestica and vombatus ursinus was 67%, 100%, 100%, 36%, 100%, 100%, 67%, 100%, 100%, 92%, 99%, 99% and 99%. In the phylogenetic tress constructed with the forteen 18S rRNA by Treeview, the Kunming mus musculus clustered with cricetulus griseus, sus scrofa and rabbit, which was nearer to cricetulus griseus and was most far away from macaca fascicularis.(3) After sencodary structure analyses of 18S rRNA of mus musculus, an oligonucleotide fragment for RNAi was designed and synthesized, which was transformed into plasmid, and restriction enzyme analyses and sequencing results should the expression plasmid pGPH1/ GFP/Neo-mouse-sh 18S rRNA were constructed for RNAi successfully.

结果①通过RT-PCR检测显示18S rRNA基因在小鼠卵巢组织和单个GV期、MⅠ期卵母细胞中均有表达，且在未成熟卵母细胞中，MⅠ期的表达明显强于GV期的表达；②RT-PCR产物克隆测序结果显示：昆明小鼠18S rRNA基因保守区序列与基因库序列[NR_003278保守区部分(791bp～1153bp)]完全一致；Blastn比对结果发现：在不同物种中差异较小，选出14种生物18S rRNA全序列经VECTOR NIT 9.0软件分析，提示昆明小鼠18SrRNA与牛、人类、刺猬、中国仓鼠、猪、犰狳、褐鼠、兔子、马、食蟹猴、负鼠、短尾猊、袋熊的18S rRNA的相似率依次为67%，100%，100%，36%，100%，100%，67%，100%，100%，92%，99%，99%，99%；Clustal 1.81和Treeview构建出的分子进化树表明：在上述14种生物中昆明小鼠与中国仓鼠进化关系最近，与兔子、猪聚成一簇，与食蟹猴进化关系最远；③根据18S rRNA二级结构设计并合成RNA干扰寡核苷酸片段，重组质粒经过限制性内切酶及测序表明成功构建了pGPH1/GFP/Neo-mouse-sh 18S rRNA干扰表达质粒。

The signals look like noncontinuous beads on the DNA fiber,which consist of multi-copy and arrange tandemly.Fiber-FISH results with 45S rDNA probe showed that an average signal length is about 3～11μm in many tandem copy sequence(measure number,n=8),so we estimated the size of each copy to be approximately 11～30kb in cotton genome.There are dual signals on each end of the mitosis metaphase chromosomes and pachytene chromosomes hybridized with telomere DNA probe.The signals also look like non-continuous beads on the DNA fiber,the length is about 1～9μm,so we estimated the size is about 4～27kb.The signals almost covered the whole mitosis metaphase chromosomes and pachytene chromosomes of G.arboretum Shixiya 1 hybridized with gDNA probe,the euchromatin zone and heterochromatin zone were identified clearly on pachytene chromosomes,and the signals also look like non-continuous beads.Two BAC clones 150-D-24 and 182-F-9 in DNA BAC library of Pima90 were selected as probe to hybridize with mitosis metaphase chromosomes,pachytene chromosomes and DNA fiber of G.raimondii.

棉花FISH技术系统的应用研究。45S rDNA在亚洲棉石系亚1号中期和粗线期染色体上有两对大的信号，在DNA纤维上信号为非连续的念珠状，多个拷贝串联排列，每个拷贝长度大约在3～11μm，推测每个拷贝的实际长度为11～30kb；端粒序列在亚洲棉石系亚1号中期和粗线期染色体端部都有双点信号，在DNA纤维上信号也为非连续的念珠状，不同染色体上信号长度大约在1～9μm不等，推测实际长度为4～27kb；gDNA信号几乎布满整个亚洲棉石系亚1号中期和粗线期染色体，在粗线期染色体上能明显区分出常染色质区和异染色质区，DNA纤维上的信号均为非连续的念珠状。

结果 正常滑膜皱襞和病理性滑膜皱襞在滑膜细胞增生及小血管增生、间质纤维化及玻璃样变、软骨化生组织学改变方面，差异均有显著性（P＜0.01）；IL-1、IL-6在病理性滑膜皱襞内的增生滑膜细胞、单核及淋巴细胞和在正常滑膜皱襞内的表达差异均有显著性（P＜0.01）； MMP-1、TIMP-1在病理性滑膜皱襞和正常皱襞内的阳性表达，差异具有显著性（P＜0.01，P＜0.05），MMP-1在增生滑膜衬里层细胞、单核和纤维母细胞、血管内皮细胞和软骨化生的软骨细胞呈阳性表达；TIMP-1只在滑膜衬里层细胞和少量纤维母细胞有表达。

A competitive ELISA kit for detect ENR was developed with ENR mAb and its traits were tested.Two hybridoma lines were filtered and the best one was 4G1-B3,which secreted ENR mAb with indirect ELISA titers of 1∶1.024×10~6 in ascites.Isotype of the two mAb was IgG_1.ENR mAb had a high affinity constant with 9×10~(10)L/mol.The ENR-Kit had the linear detection range of 0.05～101.6μg/L,the sensitivity of 0.05 μg/L and a good sensitivity with an IC_(50) of 1.1μg/L to ENR,cross-reactivity to other compounds less than 0.01%.The average recoveres of ENR spiked in chicken,liver,fish and milk were 81.5%,87.6%,84.3% and 95% respectively.The coefficient variation was below 10%.

结果表明：筛选出的2株杂交瘤细胞，单抗亚类均为IgG1型，其中4G1-B3株的ENR mAb间接ELISA效价为1：1.024×106，亲和常数为9×1010L/mol；竞争ELISA试剂盒的线性检测范围为0.05~101.6μg/L、灵敏度0.05μg/L、半数抑制浓度(IC50)1.1μg/L，与其他化合物的交叉反应率均小于0.01%，鸡肉样、鸡肝样、鱼肉样和牛奶样的平均添加回收率分别为81.5%8、7.6%、84.3%和95%，不同基质添加恩诺沙星标准品做竞争ELISA对ENR-Kit检测结果影响小，试剂盒保存期6个月以上。
