Theurinary IL-6 level positively correlated with density of glomerular matrix membrane, global sclerosis, fiber or fibrocellular crescents and interstitial fibrosis (p. 05). According to the degree of density of glomerular matrix membrane and interstitial fibrosis, urinary Col-IV level had better correlation than urinary TGF-betal and IL-6 levels.In IgAN, Col-IV showed increased expression in diseased renal tissue whereas the site of expression of TGF-betal was mainly localized within the cytoplasm of tubular epithelial cells. Interstial expressionwas also present but glomerular TGF-betal expression was found only in patients with heavy mesangial proliferation. There was a significant correlation between glomerular positivity for Col-IV and severity of histological damage. There was also a significant correlation between positivity for TGF-betal and Col-IV in the tubular epithelial and interstitial lesions. In contrast, there was no ralationship between glomerular positivity for TGF-betal and severity of histological damage.The urinary TGF-betal level paralleled tubular TGF-betal expression.

结果 ①IgAN患者尿TGF-β1、IL-6、Col-Ⅳ水平较健康人明显增高(P＜0.01)，该变化与血中的浓度无关(P＞0.05)；②尿TGF-β1水平与小管间质TGF-β1阳性表达呈正相关(P=0.000)，而与小球TGF-β1阳性表达无关(P＞0.05)，尿Col-Ⅳ水平与小球和小管间质Col-Ⅳ阳性表达均呈良好的相关性(P＜0.01)，还与小管间质TGF-β1阳性表达呈正相关(P＜0.05)：③小球Col-Ⅳ阳性表达与肾组织慢性病变密切相关(P＜0.05)，小管间质Col-Ⅳ和TGF-β1阳性表达均与肾小管间质病变呈良好的相关性(P＜0.01)，而小球TGF-β1阳性表达与肾组织损伤无关(P＞0.05)；④尿TGF-β1、Col-Ⅳ水平与肾小球基质基底膜面密度、小管间质病变呈正相关(P＜0.01)，与小球内细胞数呈负相关(P＜0.05)，该结果与其在组织中的表达一致；尿IL-6水平浙江大学硕士学位论文尿TGF一B一、IL一6和Col一IV在IgA肾病中的应用价值与基质基底膜面密度、球性硬化、纤维或细胞纤维新月体所占肾小球百分数及小管间质病变均有显著的相关性(F.05)；在轻度肾病理损伤时，尿'l'G卜pl、I卜6、Col一IV水平即升高，而尿Col一W在反映细胞外基质积聚和间质纤维化程度上比尿TGF一pl和IL一6有更好的相关性。

The increased amount of non--exchangeable NH4^＋-N during the incubation indicated as the following tendency, namely, Eum-Orthic Anthrosols 〉 Los - Orthic Entisols 〉 Hap-Ustic Isohumisols 〉 Ust-Sandiic Entisols. Organic material influenced significantly the increased amount of non-exchangeable NH4^＋-N （P was 0.0002, 0.0004 and 0.0003 when incubated at 20 d, 50 d and 60 d, respectively）. NH4^＋-N increased remarkably when Stlpa bungeana and Medicago sativa were added compared with no addition. The contents of non-exchangeable NH4^＋-N increased significantly when added （NH4）2SO4 compared without addition at 20 d, 40 d and 60d of incubation and P was 0.0037, 0.0033 and 0.0027, respectively. It was the result that the NH4^＋-N from （NH4）2 SO4 was fixed within the soil. The increased amount of non-exchangeable NH4^＋-N increased significantly for different soils, on which different vegetation types grew before the collection of soil samples, when incubated at 20 d （P=0.0434）, but not significantly at 40 d and 60d （P=0.7378 and 0.5375）. The increased amount of non-exchangeable NH4^＋-N in the soil, on which crop straw and nitrogen fertilizer had been incorporated for a long-term period, was larger than that of no addition, but it was not significantly different among these two fertilization models. Soil clay, total N and organic matter were positively correlated remarkably with the contents of non-exchangeable NH4^＋-N, the increased amount of non-exchangeable NH4^＋-N had no correlation with soil clay, whereas it had significantly positive correlation with total N and organic matter.

培养期间非交换性NH4^＋-N的增加量均表现为以土垫旱耕人为土最大，其次是黄土正常新成土，简育干润均腐土和干湿砂质新成土较小；添加有机物料极显著影响培养期间的非交换性NH4^＋-N增加量（培养20d、40d和60d时P分别为0.0002，0.004和0.0003），表现为紫花苜蓿和长芒草土壤非交换性NH4^＋-N的增加量均极显著高于不添加有机物料的对照土壤；在培养20d、40d和60d时，加（NH4）2SO4土样非交换性NH4^＋-N的增加量显著大于不加（NH4）2SO4土样（户分别为0.0037，0.0033和0.0027），这是土壤对（NH4）2SO4中NH4^＋-N固定的必然结果；不同植被类型土壤培养20d时的非交换性NH4^＋-N增加量差异显著（P=0.0434），培养40d和60d时差异不显著（p分别为0.7378和0.5375）；长期秸秆和氮肥配施土壤非交换性NH4^＋-N增加量大于不施肥对照土壤，但差异不显著土壤黏粒、全氮和有机质与培养0d、20d、40d和60d时土壤的非交换性NH4^＋-N含量均呈极显著正相关；而非交换性NH4^＋-N的增加量与粘粒无相关性，但与全氛和有机质呈显著正相关。

The chromatin unfolding assay showed that ,like the wild-type transactivation domain, two variants that represent benign polymorphisms did not induce chromatin unfolding or only induced subtle change. Contrary to the behaviors of the wild type and two benign variants, four cancer-predisposing mutations in the transactivation domain superactivate the chromatin unfolding. The results suggest that the chromatin unfolding assay can aid in the characterization of deleterious mutations in the C-terminal transactivation domain of BRCA1 and may provide more reliable presymptomatic risk assessment.

Results were shown as followings:(1) Sodium selenite at 0～2.5 μmol/L significantly increased the antioxidative capacity of L-02 cells without having remarkable impact on SMMC-7721 cells;(2) Sodium selenite at concentrations above significantly increased telomerase activity, hTERT gene expression and telomere length of L-02 cells without significant impact on SMMC-7721 cells;(3) Sodium selenite at higher concentrations (larger than 5 μmol/L) resulted in peroxidation of L-02 cells, while scutellarin significantly counteracted its effect;(4) Selenium-rich amino acids from silkworm pupas in the range of 0.5～2.5 μmol/L Se significantly inhibited SMMC-7721 cell growth, induced apoptosis and cell cycle change, and the generation of reactive oxygen species. In contrast, sodium selenite and selenomethionine only had weak impact on them at the same concentrations;(5) A new selenium-containing protein was found from selenium-rich silkworm pupas, which is worthy to be study further;(6) An expression vector containing ansense RNA of hTERT gene were constructed and used to transfect SMMC-7721 cells. They were observed to inhibit hepatoma cells.

结果如下：（1）0～2.5μmol/L亚硒酸钠显著性增强L-02细胞的抗氧化能力；而对SMMC-7721细胞的作用不显著；（2）该浓度硒显著性提高L-02细胞端粒酶活性、增强hTERT基因表达和延长细胞端粒长度；而对SMMC-7721细胞的作用均不显著；（3）高浓度硒（5μmol/L以上）显著性抑制L-02细胞生长、致细胞过氧化，灯盏花素能拮抗硒所致过氧化、降低硒毒性；（4）0.5～2.5μmol/L富硒蚕蛹氨基酸显著性抑制肝癌细胞SMMC-7721生长、导致细胞凋亡和周期改变、诱导细胞产生活性氧，同浓度亚硒酸钠和硒代蛋氨酸对其抑制不显著；（5）富硒蚕蛹蛋白经分离纯化和鉴定后发现存在一新含硒蛋白，其结构和功能有待研究；（6）通过已有的含hTERT基因质粒，成功构建hTERT反义RNA表达质粒，转染SMMC-7721细胞后对其生长具有抑制作用。
