A independent life individuality is finally formated from singlecell ferti1ized egg throngh egg cleavage、morula stage、blastocoele stage、protogaster stage and orange etc, and concerned in major basal questions of development and gene expression.

单细胞的受精卵通过卵裂、桑椹胚、囊胚、原肠胚及器官的发生等，最后形成一个能独立生活的个体，涉及发育和基因表达的许多基本问题。

The results showed:(1) the cellule morphous and the disposition in chick embryo of PGCs were coincident no matter stained by PAS or HE staining, and HE staining could disclose the morphologic characteristics more clearly, exactly and completely;(2) genesis of blood island could be observed at the boundary of light and dark region of the extraembryonic blastoderm at about 26 hours;(3) both the blood vessel endothelium cells and free cells of the blood island were differentiated from PGCs. The generating of genuine yolk sac was at about 44-48 hours.

结果表明：①用经典的识别原生殖细胞的PAS染色方法与HE染色所观察到的PGC在鸡胚的分布及细胞形态基本一致，而HE染色较PAS法能更有助于清晰、完整、准确地识别PGC的细胞内部结构特征；②在孵化26小时的鸡胚胚外胚盘的明暗交界处可见血岛的发生，血岛发生的时间比真正的卵黄囊产生时间（大约44-48小时）要早很多；③血岛周围的血管内皮细胞与管腔中游离细胞都来自PGC。

Result The embryonic development of Silnrus asotus in Dongting Lake was divided into 7 stages, namely, blastoderm stage, cleavage stage, blastula stage, gastrula stage, neurula stage, organogenesis stage and pre-hatching stage.

GV oocytes with normal shape were not obtained in the group which was stored at 20℃ for 24 h and at 25℃ for 14 h; A blastula rates (64%) of all groups in average was gotten in culturing 2-cell in vitro, and there is no significant difference in temperature and time of storage. After transplantating some of the 2-cell embryos which were obtained from GV oocytes stored at 15'C for 8 h, normal infant mice could be obtained.

在20℃下保存24h和25℃下保存14h时，难以获得形态正常的GV期卵；体外受精获得的2细胞期胚经体外培养，总体上有64%的胚胎发育至扩张囊胚，未见有保存温度和保存时间的显著影响，且利用在15℃保存8h后的GV卵获得2细胞期胚的移植获得正常新生小鼠。

The aim of the present study was to isolate and culture ESCs and EGCs from goat embryos and fetuses of different development stage, and to compare some factors influencing the efficiency of establishing those stem cell lines by adopting the methods of mouse and human. The results obtained were as follows: 1. A total of 46 Guanzhong dairy goats were superovulated with a result of total 307 embryos including 39 embryos of 4-cell to 16-cell, 128 morulae, 89 blastocysts and 51 hatched blastocysts.
