We microinjected one of the double positive ES cell clone into the mouse blastocysts (3.5days), cultured for 4-5 hours, then implanted the blastocysts in the uteri of the pseudopregnant mice.

选择其中的一个多能胚胎干细胞显微注射到孕3.5天的小鼠囊胚内，培养4-5小时后，将囊胚移植入假孕小鼠的子宫中。

MATERIALS AND METHODS: Arg to Ser mutation was introduced into the 249 position of the p53 gene by knock-in method. These ES cells with this mutation were selected according to the homologues-recombination with PCR and Southern blot. The positive ES cells without a selection marker were injected into blastocysts recovered from Hprt(superscript -/-) mice, which were derived from Hprt-deficient ES cells. The injected blastocysts then were implanted into pseudopregnant females.

材料与方法：利用基因打靶技术在小鼠胚胎干细胞p53基因249编码子中引入点突变，使编码子249由精氨酸变成丝氨酸，然后将含突变的ES细胞显微注射到Hprt小鼠囊胚中，将注射过的囊胚植入假孕的雌性小鼠子宫，到第14d取小鼠胚胎纤维母细胞，用含HAT的培养液筛选出从ES细胞分化而成的鼠EF细胞，经测序证实细胞含有由249Arg到Ser的突变。

By non-enhancement scan, 12 cases showed low density mass, 4 cases equal density mass, 1 case slightly high density mass, and 2 cases stippled calcification mass. By enhancement scan, 13 cases showed inhomogenous enhancement in cortical phase, 2 cases showed slight to moderate enhancement. Another 2 cases of small cystic RCC presented enhancement of wall with nodules and the partition. The enhancement of all tumor cases faded away in the following parenchymal phase and pyelographic phase, demonstrating some degrees of "fast washing out" phenomenon; 4 cases presented with pseudocapsule. The reconstruction of 17 "MPR/MIP" and 9 "VR" showed clearly the relations of kidney vessels, tumors and neighbor tissues and accessory renal arteries were founded in 2 cases.

结果：17例中小实性肾癌15例，小囊性肾癌2例；CT平扫呈低密度12例，等密度4例，稍高密度1例，伴有斑点状钙化2例；增强扫描皮质期13例呈不均匀明显强化，2例呈轻中度强化，另2例小囊性肾癌的囊壁、壁结节及囊内分隔明显强化；实质期、肾盂期扫描所有肿瘤强化均迅速减退，呈&快进快退&改变；4例可见假包膜。17例MPR/MIP、9例VR重建均能清楚显示肾血管、肿瘤与邻近组织的关系，并发现副肾动脉2例。

The activty of SOD in vesicle solution is then determined. But still no apparent superactivty is found due to the following reasons: 1. 研究结论：(1)与VSMC直接接触可以诱导MSC向血管成分细胞分化；(2)构建的Dox-on可调控的MSC细胞株诱导活性可靠，使外源AT2R基因的表达处于有效的主动控制下；(3)局部导入MSC不影响球囊损伤后大鼠颈动脉新生内膜增生；(4)由MSC介导的AT2R可调控表达受到Dox的良好调控，可有效抑制球囊损伤后大鼠颈动脉新生内膜增生；(5)AT2R影响血管损伤后修复时ECM成分的表达，并促进细胞凋亡的发生，可能是AT2R介导抑制新生内膜形成的机制。
