And abortion rate,efflux time of gestation sac,time of colporrhagia and returning menstruation, adverse reactions were examined.results the abortion rate,efflux time of gestation sac,time of colporrhagia and returning menstruation in three groups didn't have significant difference.but adverse reactions in them were shown significant difference.the complete abortion rate of pregnancy women less than 6 weeks was higher than that of 6～8 weeks.conclusion shorter course of treatment,minor response were shown.group a possesses advantages of short time,exact effect and little side effect,especially in less than 6 weeks duration of pregnancy.

结果：三组完全流产率、孕囊排出时间、出血时间及月经复潮时间差异无显著性；a组妊娠小于6周者完全流产率与妊娠6周～8周者相比差异有显著性；疗程短者出现副反应小，三组差异均有显著性。结论：米非司酮服用时间越短，其胃肠反应与头晕乏力等副作用越小；a组具有用时短、疗效确切、副作用小等优点，尤其对孕周小于6周患者流产效果好。

By using immunohistochemistry, RT-PCR and Western-blot, the content of this research included:(1) The expression and localization of COX-2 in mature rat and human testis were examined.(2) We investigated the changes of spermatogenesis and testosterone levels in testis after by using the COX-2 specific inhibitor-rofecoxib at 2, 4 and 8 weeks.(3) To explore the relationship between the intratesticular level of testosterone and expression of COX-2 by completely destroying the Leydig cells of mature male rats with injection of a single I.

本课题应用免疫组织化学、RT-PCR和Western-blot等方法检测以下内容：(1)在mRNA和蛋白水平检测正常成年男性及大鼠睾丸组织中COX-2的表达及定位情况；(2)实验组大鼠持续给予COX-2酶的特异性抑制剂Rofecoxib，分别于用药后2周、4周和8周检测大鼠的精子生成以及睾丸组织内睾酮浓度的变化；(3)另一组实验大鼠给予特异性Leydig细胞灭活剂EDS，在特定时间内抑制睾丸组织内合成的睾酮，于用药后2周和4周，采用免疫组织化学及RT-PCR方法观察大鼠睾丸组织内睾酮浓度变化对COX-2合成的影响。

Take the blood specimen to measure the hepatic and renal function and the tissue biopsy after the active and motive abnormities called toxical manifestation such as posture changes, abnormal sound, uneasiness, quiet, thrill, ataxia, convulsion and so on with toxic dose. Measure the tissue biopsy after death with lethiferous dose. If no toxicity or death occur after the experiment of 12 weeks, measure the hepatic and renal function and the tissue biopsy. There was no rat which had the toxical manifestation such as active or motive abnormities and 3 rats died because of environment or others after the experiment of 12 weeks.

实验中分别测定治疗剂量组小鼠在用药4周，8周，12周后的肝肾功能，并进行组织病理检查；中毒剂量组小鼠在出现行为、动作异常，如改变姿势、叫声异常、不安、安静、震颤、运动失调、惊厥等中毒表现时，取血液标本测定肝肾功能，并进行组织病理检查；致死剂量组小鼠发生死亡后进行组织病理检查；若12周实验结束时无小鼠出现中毒表现或死亡，则取血液标本测定肝肾功能，并进行组织病理检查。12周实验结束时，无小鼠出现动作、行为异常等中毒表现，有3只小鼠因为环境或其它原因发生死亡，病理检查结果正常。

In RCS rats, amplitude of a-wave in scotopic rod response and photopic response after 4-week transplantation was increased (PO.05); while latent period of a-wave in scotopic rod response after 4-week transplantation was shorten (PO.05).Conclusion 1. At the age of 3 months, photoreceptors of the RCS rat degenerated completely, which leaded to the reduction of amplitudes of a-wave and b-wave.
