Experiments on decalcification and activation of osteoinductive ability, comparison between the osteogenesis of demineralized bone matrix in orthotopic and in heterotopic sites, and correlation between immune response to allograft of bone and osteoinductive activity of the graft, were reported in this paper. 1. Syngeneic deminerolized bone matrix and the mixture of DBM with bone ash or acid-soluble protein extracted from demineralization was implanted into muscle pouch of mice, subsequently, the inhibition of DBM osteoinduction by bone ash or acid-soluble protein was demonstrated with histological and by means of alkaline phosphatase assay.

本文对脱钙处理使骨诱导活性表达的作用、脱钙骨基质植入不同部位的成骨表现，以及同种异体骨移植引发的免疫反应与骨诱导活性的内在关系三方面进行了研究：（1）在小鼠肌肉植入同系脱钙骨基质及其与脱钙所去除的骨矿盐和酸溶性蛋白混合物，碱性磷酸酶和组织学检查表明：骨矿盐和酸溶性蛋白具有抑制脱钙骨基质骨诱导活性的作用。

The they and blood fat level in blood plasmaand expression of MCP-lmRNA, VCAM-1, MMP-2 of aorta on intervention group aremuch lower than that of model group (P＜0.05); Bcl-2 of intervention group isstrengthened, and the pathomorphology and immunohistochemistry of aorta showpathological changes are slighter than model group.

结果：模型组，血浆中同型半胱氨酸浓度、血脂水平、主动脉MCP-1mRNA表达显著高于正常对照组(P＜0.05)，免疫组化示主动脉内皮细胞VCAM-1、MMP-2表达增强，Bcl-2表达降低；主动脉切片发现动脉内膜不连续，部分剥脱，平滑肌细胞排列疏松、紊乱，主动脉壁均见到程度不同的脂质浸润，其病变弥漫，泡沫细胞较多，可见斑块突出管腔；干预组，血浆中同型半胱氨酸含量、血脂水平显著低于对照组和模型组(P＜0.05)，主动脉MCP-1mRNA、VCAM-1、MMP-2表达显著弱于对照组和模型组(P＜0.05)，Bcl-2表达增强(P＜0.05)，主动脉病理形态学和免疫组织化学检测示病变轻于模型组。

In every group, rabbits were subdivided into experimental and control subgroups. 2 Rabbits were bullets injected followed by continuous injected with 13C labeled leucine, glucose, and lactic acid; 3 Blood were drewed before and 150, 160, 170, and 180 min after the initiation of isotopes injection for material analysis; 4 Exhale gas were collected every 5 min in the first 30 min followed by every 30 min there after for material analysis; 5 After centrifuge in low temp the supernatant of the blood samples were collected and went through axon and anon exchange column treatment; 6 Treated blood sample was used for 13C labeled leucine, glucose lactic acid examination through mass spectrograph; 7 The exhale gas was collected for 13CO2 exam through gas-phase mass spectrograph; 8 CO2 total production rate (V13CO2), body various substances production, oxidation speed, and substances metabolic percentage all can be calculated through equations provided by references below; 9 Data was processed through student t test.

分亮氨酸、葡萄糖和乳酸三组各取健康新西兰兔16只，每组再分为高代谢脓毒症组和对照组两部分，建模方法同第一部分；2）三组分别先静脉弹丸式推注再持续灌注13C标记的亮氨酸、葡萄糖和乳酸；3）灌注前抽取动脉血作为背景值，灌注达150，160，170和180min分别抽取动脉血2ml用于质谱分析；4）实验开始后前30min中每5min收集呼出气一次，随后每30min收集呼出气一次，用于气相质谱分析；5）血样经过低温离心取上清液，过阳离子及阴离子交换树脂，再经衍生化处理；6）处理过的血样进入气相色谱－质谱仪，测量其13C标记的亮氨酸、葡萄糖和乳酸丰度；7）实验兔的呼出气通过13CO2气相质谱分析仪测定其中的13CO2丰度（E13CO2）；8）利用文献提供的公式算出CO2总生成率（V13CO2）以及机体各物质的通量、氧化速率以及物质代谢百分比等；9）数据分析处理采用t检验。

首先研究本区的构造特征，根据断裂的平面形态及相互切割关系，乌夏地区发育两种构造体系：北东向构造体系和近东西向构造体系，前者形成早，控制了该区的构造格局，晚期有3次近南北向挤压作用叠加；乌夏地区南北根据构造变形的强度不同分为冲断带、断褶带、单斜带，东西由于横向调节断层的存在分为乌尔禾断褶区、乌-夏冲断区和夏-红断褶区，进一步划分为九个次级单元；构造调节带与储集层的分布关系密切，通常是大量富砂质沉积的源区，乌夏地区发育三种类型的构造调节带：同向接近型、同向叠覆型和同向共线型。

The PSCA_3 fragment was selected for its superior expression level in eukaryotic cells.Then the sig-PSCA_3-Fc-GPI genetic fragment was cloned into pVAX1-neo-IRES-GM/B7 vector to construct the final immunological inhanced DNA vaccine pVAX1-PSCA_3-FcGB. Immunofluorescence and flow cytometry were used to confirm the expression of PSCA_3 fragment by transfected into Cos7 cell.Finally,the anti-tumor effect of pVAX1-PSCA_3-FcGB was tested in murine prostate cancer model generated by RM-1 cell line.The animal was immunized with pVAX1-PSCA_3-FcGB DNA vaccine by intramuscular injection plus electroporation,pVAX1 and pVAX1-PSCA_1-FcGB plasmid were used as control.The inhibitory effect of tumor was investigated by observion of forming time,volume and inhibition ratio of tumor.Results:DNA sequencing conformed that the heterological PSCA fusion antigen fragment which was synchronized by overlapping-extending-PCR,was consistent to design.Enzyme digestion analysis showed that the 1 to 4 copies heterological PSCA fusion antigen fragments were constructed successfully.

方法(1)检索GenBank，选择包含人主要T细胞抗原表位序列的人PSCA基因片段，应用异种化抗原设计技术，保留人T细胞抗原表位，设计异种化PSCA融合抗原片段；(2)根据核酸序列按中心模板法设计引物，应用重叠延伸PCR技术拼接合成异种化PSCA融合抗原片段基因，以PCR、限制性酶切和DNA序列测定法进行鉴定：(3)利用DNA限制性内切酶BssHⅡ和MluⅠ酶切后粘端互补的特点，采用同尾酶法构建1—4拷贝异种化PSCA融合抗原片段(PNCA_1-PSCA_4)，并将上述片段分别插入真核表达载体pCI-neo-Fc-GPI中，转染293T细胞，借助免疫荧光+流式细胞术考察插入片段表达效率，最终选定PSCA_3片段进行下一步研究；(4)将sig-PSCA_3-Fc-GPI基因片段自pCI-PSCA_3-Fc-GPI质粒上切下，插入pVAX1-neo-IRES—GM/B7载体中，构建免疫增效DNA疫苗pVAX1-PSCA_3-FcGB，并应用转染Cos7细胞+免疫荧光/流式细胞术方法鉴定其在真核细胞中的表达情况；(5)给8周龄雄性C57BL/6小鼠皮下种植RM-1细胞，制备小鼠前列腺癌模型，并采用股四头肌肌肉注射+电脉冲法(Electroporation，EP)接种DNA疫苗质粒pVAX1-PSCA_3-FcGB，同时接种pVAX1空载体质粒和pVAX1-PSCA_1-FcGB质粒作为对照，通过观察计算免疫动物的成瘤时间、肿瘤体积和抑瘤率，来评价该DNA疫苗在小鼠体内的抑瘤效果。
