①Rat MSC and VSMC were cultured and identified, respectively. MSC were labeled with DAPI firstly, and then co-cultivated with VSMC. The changes of morphology and ultrastructure of co-cultured cells were observed. Immunfluorescence analysis was performed by using monoclonal antibodies against specific antigen.②We established the regulatable system in two steps: a stable MSC line expressing rtTA has been constructed and characterized firstly by transfected with pUHD 17-1hyg and then selected by hygromycin B; in a second step, this line was used for trandfer the AT2R gene to MSC to get the well establishing double stable MSC lines;③The expression of AT2R regulated by doxycycline was evaluated by western blot;④The MSCs were transduced into rat carotid arteries with regulatable AT2R gene after the establishment of rat carotid balloon injury restenosis model. The intimal/medial area ratio were measured by digital analysis system.

研究方法：(1)密度梯度离心法及胶原酶消化法分别培养原代大鼠MSC及VSMC，细胞共培养并行免疫荧光化学染色和透射电镜观察超微结构；(2)组成受Dox调控的哺乳动物表达系统的四种成分的转化、扩增及提纯并酶切鉴定；(3)采用常规分子生物学方法连续两个回合转染体外培养的MSC，并分别采用发光计检测不同细胞克隆萤光素酶活性改变以及RT-PCR方法检测AT2R目的基因mRNA表达情况，根据各个细胞克隆受Dox调控表达的程度，选择低背景、高诱导表达AT2R的细胞系，作为双重稳定MSC细胞系；蛋白免疫印迹法观察该细胞系在Dox调控下AT2R表达的时相性、持续性及在不同浓度Dox调控下的表达情况；(4)建立大鼠颈动脉球囊损伤动物模型，将双重稳定MSC在术中导入血管，分别于14 d、28 d进行病理切片，检测可调控AT2R对新生内膜增生的影响；采用RT-PCR免疫组织化学免疫荧光等技术观察AT2R基因在新生内膜中的表达以及细胞外基质成分表达的改变，TUNEL法检测血管组织中细胞凋亡的变化情况。

In summary, the BRD7 gene acted as a candidate of tumor suppressor gene with NPC. The overexpression of BRD7 can partly reverse malignant phenotype of NPC cell line. The suppression effect of BRD7 on NPC tumorigenesis my be achieved by recognizing acetylated histone peptide through their motif-bromodomain, then modulating gene transcription by taking part in histone acetylating and chromosome remodeling, finally influencing signal-transduction pathways.

综上所述，BRD7基因作为一个重要的鼻咽癌抑瘤基因侯选者，在鼻咽癌细胞中的过表达后，可导致鼻咽癌细胞 HNE蛋白质表达谱发生改变，逆转其恶性表型，其作用机理可能是：BRD7基因通过其功能域彭聪硕士学位论文10Bromodomain与乙酚化的组蛋白特异性结合，参于染色体的乙酚化，染色质的组装，从而影响基因转录的调控，最终影响细胞内的信号传导通路并实现对细胞周期的调控，从而发挥抑制鼻咽癌细胞生长的作用。

In this experiment, a new method was used to purify the toxin protein produced by Verticillium dahliae. The supernatant of fungus culture was frozen and dried with Lyophilizer first, and then dialysed by Dialysis Membranes (MWCO 1000) after dissolved in distilled water. This method can eliminate the salt and sucrose in culture medium and reserve the protein component almost completely. VD-toxin were analysed using sodium dodecyl sulfate polyacrylamide gel analysis. The results indicated that the protein components were very complex, and included glycoprotein within 35.8kDa-83.2kDa. Furthermore, treatments on glycoproteins with high temperature, conA and proteinase alleviated, not abolished, their activities.NO and H_2O_2 production were assayed in two cultivars of cotton cells which were treated with VD-toxin.

本实验首先确定了用冷冻浓缩后透析的方法初步提纯棉花黄萎病菌毒素，该方法能有效去除粗提毒素中的蔗糖和盐离子并能最大程度的保留黄萎病菌分泌的蛋白成分；对毒素蛋白成分鉴定结果表明蛋白条带多，每种蛋白的含量少，小分子量蛋白含量多，糖蛋白染色结果显示，毒素蛋白中分子量在35.8kDa-83.2kDa之间的蛋白大多数为糖蛋白，分子量小于35.8kDa的蛋白多为非糖蛋白；性质鉴定结果显示，毒素中有活性的蛋白成分具有部分可耐高温高压，且毒素蛋白中的蛋白成分和糖基成分都具有致萎活性。
