The feasibility of determining the pentosan in pre-extraction liquor from eucalyptus chips by using Douglas colorimetric method was discussed in this paper. The visible spectrums of xylose, glucose, mannose, galactose and kraft lignin were compared under the same color reaction conditions. The results showed that the disturbance from hexoses and lignin can be eliminated when the pentosan is identified and determined by the maximal absorption at 553nm.
