Based on the above-mentioned, two kinds of target glycoclusters with different glycoterminus, flexible linkers and different scaffolds were designed, disaccharide glycocluster (including TM1-TM7) and trisaccharide glycocluster (TM8-TM9) respectively, with the purpose of obtaining glycoclusters with high binding affinity to anti-Gal antibody and thus better inhibiting the HAR.

基于上述，针对HAR产生的机制，以人体中天然存在的anti-α-Gal抗体及B细胞表面的相应受体为靶点，以α-Gal抗原末端的二糖Galα1→3Gal和三糖Galα1→3Galβ1-4GluNAc片段为糖基部分，采用柔性的连接臂（氨基保护的2-乙醇胺和3-巯基丙酸）和刚性的芳香骨架及柔性的脂肪链骨架，设计了两类结构新颖的糖簇分子：即氧苷和硫苷键连接的二糖糖簇TM1-TM7）和三糖糖簇（TM8-TM9），目的在于寻找与anti-Gal抗体及B细胞相应受体有较高结合力的糖簇分子，以便有效地抑制HAR反应的发生。

Of the 8 cases, 2 cases showed blood Tb-Ab positive, 1 case was positive to antiacid bacillus of sputum, the rest cases with negative TBAb. 3 patients with 2 showed organs infection, 6 patients passed the tissue biopsy diagnose, body temperature of 6 cases fall to normal after 3-7 days of antituberculotic treatment, mild liver dysfunction appeared in 2 patients, hyperuricemia occurred in 7 patients, in addition, 1 patient developed reject reaction. Among all the cases, 7 cases healed, 1 died of pulmonary fungal infection.

其中2例血Tb-Ab，1例腹水结核抗体阳性，1例痰抗酸杆菌阳性，其余结核抗体阴性，3例患者表现为2个脏器感染，6例通过病理组织活检诊断，6例发热者在抗结核治疗3~7d体温降至正常，2例患者在治疗过程中出现轻度肝功能异常，7例出现高尿酸血症，1例出现移植肾排斥反应。8例患者7例痊愈，1例肺结核患者死于肺部真菌感染。6例发热者在抗结核治疗3~7d体温降至正常。

Objective Establishment of a hybridoma cell line secreting a monoclonal antibody to facilitate iˉdentification of human tissue kallikrein（KLK1gene,hK1protein）.Methods Mice were immunized with E.coli-exˉpressed GST-hK1fusion protein and their spleen cells were fused with SP2/0myeloblastoma cells.Specificity of the monoclonal antibody was shown by Western blotting and by immunofluorescence.Results The monoclonal antibody reacted specifically to E.coli-expressed hK1and with the KLK1cDNA-transfected COS-1cells.
