In clinical. acetylcholinesterase inhibitors reduce the breakdown of synaptic acetylcholine, have been modestly effective in improving the cognitive deficits of Alzheimer's disease.The goal of this work is to determine enzyme kinetics and mechanisms of acetylcholinesterase and butyrlcholinesterase inhibition by five cardiovascular drugs, lovastatin, simvastatin, amlodipine besylate, nifedipine, and hydralazine hydrochloride, and two benzodiazepines, diazepam and chlordiazepoxide hydrochloride. All drugs in this study are reversible mixed-type inhibitors of acetylcholinesterase and butyrylcholinesterase. The pKi values for acetylcholinesterase and butyrylcholinesterase inhibition by the cardiovascular drugs are linearly correlated with the molecular weights of the drugs with the slopes of 0.005 and 0.0021, respectively. Therefore, van der Waals' interactions between acetylcholinesterase and the cardiovascular drugs are stronger than those between butyrylcholinesterase and the drugs. This is probably due to a smaller active site gorge and a more significant peripheral anionic substrate binding site of acetylcholinesterase than those of butyrylcholinesterase.
本研究之目的系决定乙醯胆碱酯酵素和丁醯胆碱酯酵素被五种心脏血管药物lovastatin, simvastatin, amlodipine besylate, nifedipine、hydralazine hydrochloride和两种benzodiazepines:diazepam和Chlordiazepoxide hydrochloride的抑制作用之动力学及机转,这些药物都是乙醯胆碱酯酵素和丁醯胆碱酯酵素的可逆性、混合型之抑制剂,实验结果显示,五种心血管药物对於乙醯胆碱酯酵素及丁醯胆碱酯酵素抑制之pKi值和药物之分子量呈直线之关系,斜率分别为0.005及0.0021因此丁醯胆碱酯酵素,与心血管药物间之凡德瓦作用(van der Waals' )比乙醯胆碱酯酵素与心血管药物间者弱,这可能原因是丁醯胆碱酯酵素之活性区域比乙醯胆碱酯酵素之活性区域更宽广,但丁醯胆碱酯酵素之周边阴离子区域不及乙醯胆碱酯酵素之周边阴离子区域者明显重要,由於五种心血管药物对於乙醯胆碱酯酵素和丁醯胆碱酯酵素抑制之pKi值存在著线性关系表示此种抑制作用系经过共同之反应机理。

On the basis of the kinetic equation of substrate reaction in the presence of urea or guanidine hydrochloride, all microscopic kinetic constants for the free enzyme and enzyme-substrate binary and ternary complexes have been determined. The results of the present studies indicate that:①In the presence of urea or guanidine hydrochloride, enzyme-substrate complexes lose their activity less rapidly than the free enzyme. Therefore, both substrates, NADPH and 7, 8-dihydrofolate, protect dihydrofolate reductase against inactivation.②The denaturation of dihydrofolate reductase by urea follows single-phase kinetics, and changes in enzyme activity and tertiary structure proceed simultaneously in the unfolding process, so it may be an"all or none"process.③The GdnHCl-induced unfolding of the dihydrofolate shows a biphasic transition, while the change in the enzyme activity is a single exponential process. The rate constant of inactivation is consistent with that of the fast conformational change. Therefore, the kinetic intermediate of protein unfolding should be a partially folded and inactive form.
我们根据在脲或盐酸胍存在下的底物反应动力学方程求得游离酶和酶底物二元、三元复合物的微观动力学常数,结果表明:①酶-底物二元、三元复合物的失活速度明显慢于游离酶,说明两个底物二氢叶酸和NADPH对酶的失活都具有一定程度的保护作用;②在脲作用下,酶的失活和构象变化均为单指数项过程,而且酶的活力丧失和三级结构变化是同时发生的,说明二氢叶酸还原酶的脲变性可能是一个"全或无"的两态过程;③在盐酸胍作用下,酶的构象变化为两相过程,而失活则是单指数项过程,酶分子构象变化的快相速度常数与失活速度常数基本一致,因此我们认为二氢叶酸还原酶的盐酸胍变性过程中存在一个没有活力、但仍具备一定空间结构的变性中间体。

The formal potentials in cyclic voltammograms of these 3+ valence complexes are followed: Co〓-TPPS (-0.08V )>Fe〓TPPS (-0.14V)>Mn〓-TPPS (-0.23V). The electrochemical reaction of Mn〓-TPPS is reversible, and both of Co〓-TPPS and Fe〓-TPPS are quasi-reversible. The standard heterogeneous rate constants of Co〓-TPPS and Fe〓-TPPS were determined to be 1. 4×10〓cm/s and 1.4×10〓cm/s respectively. The rates of electrode reaction are as followed: Mn〓-TPPS>Co〓-TPPS>Fe〓-TPPS, e.i. Mn〓-TPPS is the most easily oxidized by the dissolved oxygen, and Fe〓-TPPS is oxidized slower than Co〓-TPPS. Ni〓-TPPS can not be oxidized. It is considered that oxygen plays an important role in the valence change of central metal atom and the rate of electron transfer in some metalloporphyrins.
