Methods:Establish the model of myocardial fibrosis in chronic stage of viral myocarditis by injecting CVB3 into Balb/c rats,divide these rats into model group、Tong xin luo low dose group、Tong xin luo medium dose group、Tong xin luo high dose group and set up normal group,after four weeks of giving medicine execute the animals and observe the ultrastructure of myocardial cell with tramission electron microscope,the quantity of myocardial collagen was observed by staining with H.E and Sirus red in supersaturated picric acid solution;the amount of HA in serum was measured with the method of radiative immunity;the quantitative expression of fibrosis factor—TGF、PDGF was measured with the method of immunity histochemistry.

腹腔注射柯萨奇B3病毒建立病毒性心肌炎慢性期心肌纤维化小鼠模型，随机分为模型对照组、通心络低剂量组、中剂量组、高剂量组，并设正常对照组，用药四周后处死动物通过透射电镜观察心肌细胞超微结构；HE染色、苦味酸天狼星红染色观察心肌胶原数量；放免法测定血清透明质酸的含量；免疫组化染色观察致纤维化细胞因子—转化生长因子和血小板衍生生长因子的定量表达。

These results suggested that the lung Fib were activated and produced excess HA, resulting in the excessive accumulation of HA within the lung at the stage of alveolitis during the process of pulmonary fibrosis. However, increased HA synthesis of lung Fib and increased HA levels in BALF merely reflect the intensity of alveolitis and the disease activity rather than the degree of pulmonary fibrosis.

以上结果提示：肺Fib在肺纤维化的早期肺泡炎阶段就处于活化状态，产生HA增多，导致肺内HA的过多聚积，肺Fib产生HA增多和BALF中HA水平的增高可反映肺泡炎的强度和疾病的活动性，但并不能反映肺胶原代谢或纤维化的程度。

The positive signal of ER and ER mRNA distributed in parenchymal and non-parenchymal hepatic cells, especially near the hepatic centrolobular and periportal areas. Ovariectomy decreased ER level and ER mRNA expression significantly (P＜0.05). Hepatic ER and ER mRNA concentration were elevated after treatment with estrogen in both ovariectomy and sham ovariectomy fibrotic groups (P＜0.05), and the elevation of ER was correlated with a marked decrease in hepatic damage and fibrosis.

Primary culture of goat mammary gland cells could be derived by outgrowth of migrating cells from fragment of tissue. Fibroblast and epithelial cells could be pured according to their different sensibility to trypsin. Complex cultures treated by 0.25% trypsin in Hanks' for 5~7min at 37℃, the dispersed cells were mainly fibroblasts,then the rest treated by 0.15% trypsin-0.02% EDTA in Hanks'for 5~8min at 37℃, the majority cells harvested were mammary epithelial cells.

用组织块培养法可获得良好的山羊乳腺细胞原代培养物；培养的山羊乳腺成纤维细胞比上皮细胞对胰蛋白酶更敏感，据此可在传代过程中将二者分离纯化，获得纯细胞系；混合培养物先用0.25%胰蛋白酶在37℃消化5~7min 所收集的细胞主要为成纤维细胞；再加0.15%胰蛋白酶-0.02鞹A 消化液在37℃继续消化5~6min 所回收的细胞绝大多数为上皮细胞，经过2~3 代，即可得到纯化的乳腺上皮细胞系。

Methods: The fibroblasts companying human hepatoma were primarily cultured with explant culture method, first passaged after they spread to the bottom of the culture bottle, and pured by enzyme digestion and repeated strapping the cells were identificated through observing the morphologic change using invert microscope and detecting expression of vimentin, keratin by iMmunohistochemical method.
