Objective: To evaluate the efficiency of sterilizing tomato seeds with 0.1% mercuric chloride and 3%-4% sodium hypochlorite solution.

评价番茄种子经0.1%升汞和3%~4%次氯酸钠溶液灭菌的效果。

Tomato seeds were firstly immersed in 0.1% mercuric chloride for 10 to 15 min with shaking and then rinsed four or five times with sterile distilled water.

另将番茄种子浸于75%酒精1~2 min，弃酒精后加入足量的0.1%升汞溶液充分振荡10~15 min，再以灭菌水充分漂洗种子4~5次。

The optimized conditions for isolation and culture of endophytic bacteria isolated from grasses in alpine area in this study. The different tissues of grasses were immersed in 0.1% of SDS for 15 min, 3% of NaClO for 3 min, 0.1% of mercuric chloride for 10 min, 75% of alcohol for 1 to 2 min. The endophytic bacteria were isolated after aseptic grinding and diluted to 10^(-3) and using TSA culture medium for 5 to 7 days under 28℃.

通过对高寒牧草内生细菌的分离培养方法研究，得出了优化的分离培养条件，即牧草不同组织器官用0.1%SDS浸泡15min、3%NaClO浸泡3min0.1%升汞浸泡10min、75%酒精1~2min处理后（各步间均用无菌水冲洗3~4次），研磨并稀释至10^(-3)，涂布于TSA培养基上，置28℃恒温条件下培养5-7d，可从高寒牧草组织中分离获得数量、种类较多的内生细菌。

Result Explants sterilized in 0.1% mercuric chloride for 8 min had the best effect. The optimum medium for callus was MS+6-BA 1.0-1.5 mg/L+NSA 0.5-1.0 mg/L.

结果］用浓度0.1%升汞溶液灭菌8 min，外植体污染率为54.55%，死亡率为3.64%；最佳诱导培养基为MS+6-BA 1.0-1.5 mg/L+NAA 0.5~1.0 mg/L。

In the end, transportation of PAHs in the environmental matrix was preliminarily investigated and discussed according to PAHs data of soils and sediments from a certain region in Shanghai. The result indicated that there are two or more pathways on PAHs transportation in the environmental matrix.

最后，对上海特定地区内PAHs在环境介质中的迁移径途进行了分析讨论，结果表明上海市土壤和沉积物等环境介质中PAHs的迁移存在多种途径。
