用球囊扩张法可成功建立大鼠颈总动脉损伤膜型；Doxy 可以显著降低血管损伤后 MMPs 活性，抑制内膜 SMCs 的增殖、新生内膜增生以及后期的管腔重构，提示它可能具有防治 PTCA 术后再狭窄的作用。

Results In the ANCA negative group, 2 cases were without malignant hypertension ,1 was acute tubular necrosis caused by herbs, 6 were scleroderma renal crisis, including 4 with renal biopsy, indicating hypertrophic arterial media, edema, thickened intima, onion- skin lesion in interlobular arteries and afferent arterioles, as well as ischemic lesion in glomeruli.

结果 11 例SSc急性肾损伤患者中，ANCA阴性9 例，6 例临床表现为硬皮病肾危象，其中4 例肾活检示小叶间动脉和入球小动脉内皮细胞增生、肿胀，内膜黏液性水肿，内弹力纤维断裂，"洋葱皮样"改变和肾小球弥漫性缺血；无恶性高血压表现2 例；急性肾小管坏死1 例。

①Rat MSC and VSMC were cultured and identified, respectively. MSC were labeled with DAPI firstly, and then co-cultivated with VSMC. The changes of morphology and ultrastructure of co-cultured cells were observed. Immunfluorescence analysis was performed by using monoclonal antibodies against specific antigen.②We established the regulatable system in two steps: a stable MSC line expressing rtTA has been constructed and characterized firstly by transfected with pUHD 17-1hyg and then selected by hygromycin B; in a second step, this line was used for trandfer the AT2R gene to MSC to get the well establishing double stable MSC lines;③The expression of AT2R regulated by doxycycline was evaluated by western blot;④The MSCs were transduced into rat carotid arteries with regulatable AT2R gene after the establishment of rat carotid balloon injury restenosis model. The intimal/medial area ratio were measured by digital analysis system.

研究方法：(1)密度梯度离心法及胶原酶消化法分别培养原代大鼠MSC及VSMC，细胞共培养并行免疫荧光化学染色和透射电镜观察超微结构；(2)组成受Dox调控的哺乳动物表达系统的四种成分的转化、扩增及提纯并酶切鉴定；(3)采用常规分子生物学方法连续两个回合转染体外培养的MSC，并分别采用发光计检测不同细胞克隆萤光素酶活性改变以及RT-PCR方法检测AT2R目的基因mRNA表达情况，根据各个细胞克隆受Dox调控表达的程度，选择低背景、高诱导表达AT2R的细胞系，作为双重稳定MSC细胞系；蛋白免疫印迹法观察该细胞系在Dox调控下AT2R表达的时相性、持续性及在不同浓度Dox调控下的表达情况；(4)建立大鼠颈动脉球囊损伤动物模型，将双重稳定MSC在术中导入血管，分别于14 d、28 d进行病理切片，检测可调控AT2R对新生内膜增生的影响；采用RT-PCR免疫组织化学免疫荧光等技术观察AT2R基因在新生内膜中的表达以及细胞外基质成分表达的改变，TUNEL法检测血管组织中细胞凋亡的变化情况。

采用兔腹主动脉内膜剥脱术加含1.5%高胆固醇的高脂饮食来建立动脉粥样硬化动物模型，并在此基础上建立兔粥样硬化腹主动脉植入支架模型，并通过组织病理学检查，来揭示支架植入后血管壁和内膜组织形态学变化和病变的发生机制。

Methods: To divide 102 patients with elderly hypertensive into 3 groups according to the difference of blood pressure result, and compare the relationship between serum uric acid and blood pressure among the different groups. They were divided into normal group、intimae tricking group、plaque shaping group and vascular stenosis group accoraing to ultrasound result. To compare the serum uric acid level and blood pressure among each group.

将102例老年高血压患者按血压值的不同分为三组，比较各组间血尿酸与血压的相关性，对各例患者分别检测血生化指标：空腹血糖、血尿酸、甘油三脂和总胆固醇，B超观察颈动脉变化，按B超结果将患者分为内膜正常组、内膜增厚组、斑块形成组和管腔狭窄组，比较各组间血压、血尿酸水平。

In the rat carotid injury model, SM16 dosed once daily orally at 15 or 30 mg/kg SM16 for 14 days caused significant inhibition of neointimal thickening and lumenal narrowing. SM16 also prevented induction of adventitial smooth muscle alpha-actin-positive myofibroblasts and the production of intimal collagen, but did not decrease the percentage of proliferative cells.
