The animal experiment stage: 40 rabbits underwent corneal epithelium curettement and limbal ablation to induce limbal stem cell deficiency and were randomly divided into four groups, simple amniotic membrane group, non-frozen cells group, glycerol group and dimethyl sulfoxide group(n=10 for each group). Searching for the activity of frozen and non-frozen cells more completely by transplanting subculturing cells and observing the accomplishment of cells function.

动物实验阶段：将40只角膜缘干细胞损伤的模型兔随机分成单纯羊膜组、未冷冻细胞组、DMSO保存细胞组和甘油保存细胞组，每组10只，用传代培养2周后的角膜缘上皮细胞作移植实验，观察细胞体外功能完成情况来进一步比较深低温保存前后角膜缘上皮细胞的活性。

The three types of reconstructed embryos(reconstructed embryo of goat-rabbit, goat-bovine and goat-goat) were produced respectively by nuclear transfer using goat ear firbroblast cells as donors and rabbit, bovine and goat ooctyes as recipients, and the factors of cytoplast environments influencing cloned embryo development in vitro, cytoplast effect in nuclear transfer, the methods of transferring cloned embryo and the effects of pregnancy factor on implantation of cloned embryo were studied in this experiment, in order to supply basis for solving the difficulty of implantation of inter-species cloned embryo in recipient and improving the efficiency of cloning.

The stereocilia of OHCs are extremely short. We can observe thephenomena of rupture and inosculation of stereocilia, which is alike senile deafness ofICR mouse. The OHCs bodies of bats have two leading characteristics. Firstly, theOHCs bodies are obviously short. The Deiters cell phalanges are of exaggerated, andthe cup formation of Deiters cell body enwrap the bottom of the OHCs.

By the test of clonal growth in soft agar medium , we compared the ability of cloning and proliferating among these three kinds of cells. The cell cycle was analyzed by FCM and morphosis of cells was observed under transmission eletron microscope.

Methods At first, retrovirus vectors encoding IFN-γ or IL-4 gene were constructed. They were transfected into PA317 packaging cells by lipofectamine, PA317IFN-γ and PA317IL-4 cells were obtained. C6 glioma cells were infected with replication deficiency retrovius containing IFN-γ or IL-4 gene, cell morphology、cell growth curve and cloning efficiency assay were examined. The intracranial C6 glioma animal model was established in immunocompetent Wistar rats. PA317IFN-γ and PA317IL-4 cells were stereotactically implanted into the tumor areas alone or together. The survival of tumor bearing rats were examined, tumor volumes were measured and immunohistochemical analyses of CD4〓 and CD8〓 T lymphocyte infiltration were performed.
