Methods:Animal models of 25 adult mixed breed dogs were established by transverse osteotomy at bilateral iliopectineal crest,which were fixed with ATMFS anterior column connector and steel plate respectively. The animals were examined at 2,4,6,8,and 12 weeks by X-ray,macropathological observation and biomechanical test. Tetracin fluorescent labeling,HE staining,Masson triad colour staining,in situ hybridization and immunohistochemical method were performed and histological image analysis were applied.

建立犬双侧骨盆弓状线骨折的动物模型，分别采用ATMFS前柱固定器和5孔重建钢板内固定，于术后2、4、6、8、12周分别行影像学检查、大体观察及生物力学测试；采用四环素荧光标记法、HE染色、Masson三色染色法、原位杂交法、免疫组织化学法进行组织学观察并图像分析；采用RT-PCR法检测骨折端不同时期骨钙蛋白和核心转录因子mRNA相对表达量的变化。

Objective: This experimental study is based on earlier animal and clinical experiment, drawn thinkings of combinations with two methods and pursued a synthesis method of high-efficiency, cheap and convenient calcium and exercise treatment. We adopt taking natural-clam's casing powder and preconcereted-intensity excises to intervene shape of osteoporosis in gelded rats, than we compare the indexes such as the biomechanics indexes of the right femur and the third lumbar vertebrae, the weight and uterus quotiety of rats before the experiment to these indexes after the experiment. We hope to find better and suitable way to prevent and treat the postmenopausal osteoporosis and offer reference for clinic.

目的 本实验的主要目的是在前期动物实验的基础上，引入两种方式联合干预的思维，立足于探求既高效又经济、方便的补钙剂加上与运动疗法相联合的方式，对切除雌性大鼠卵巢而复制成的骨质疏松症模型进行服用天然贝类蛤蜊壳粉，同时施以预定强度的运动刺激的方法，再通过试验前后大鼠体重、子宫指数的变化及大鼠股骨和椎体生物力学指标的对比变化进行分析，以期探索出防治绝经后骨质疏松症的更好的、更适宜于大众的手段；并为天然钙剂和运动联合作用防治PMOP提供参考。

The coil spring were kept constant and recorded for 0hr, 1 hr, 2 hrs, 4 hrs, 8 hrs, 12hrs, ldays, 3days or 7days. Upon completion of experiments, the maxillae were removed. The specimens were fixed in 4% paraformaldehyde in 0.1 M phosphate buffer for 24 hrs and decalcified in 10% ethylene diamine tetraacetic acid at room temperature for 5 wks.

After three months, gained blood from cardinal vein and detected the level of serum E2; put all rats to death and got femur bone tissue with asepsis condition. Detected the transcriptional level of PTHR1 gene with RT-PCR. And detected the expression level of PTHR1 protein with Western blot. Results: In ovariectomized groupthe level of serum E2 was 13.80±1.10 pmol/L, the E2 of model control group was 23.51±1.20 pmol/L.

术后3个月腹主静脉取血，测定各组大鼠血清E2水平；分别用小动物骨骼强度测定仪和双能X线骨密度仪检测右股骨生物学力学和骨密度；从左股骨中提取总RAN和总蛋白，分别检测PTHR1在转录和翻译水平上的变化。
