Based on the results of exploration and in-site test of typical waste dump in southwest mountainous area,the analysis theory and method of waste dump rainfall infiltration are discussed beginning at analysing coal waste's mechanics characteristic and seepage theoty. Fllowed estimating method of waste dump stability is discussed influenced by rainfall. At last a typical engineering example of coal waste dump landslide influenced by rainfall is researched, acquiring research fruit hereinafter.①Particular exploration of typical waste dump in chongqing area and analysis estimate to its area engineering geology are done. Granule gradation、water content、nature density、dry densit etc parameter of waste granular media are tested. The mechanics characteristic of waste are researched by in-site shear experimentation、compressibility experimentation and infiltration experimentation.②The coal waste dumping is different from other consecutive solid material as incompact granule structure. Its shear strength and transforming characteristic also have itself characteristic.
本文针对山区矸石山,结合踏勘和现场试验,从分析矸石散体的力学特性以及渗流理论出发,研究了矸石山降雨入渗的分析理论与方法,在此基础上提出了矸石山体降雨作用下的稳定性评价方法,最后进行了降雨作用下典型矸石山滑坡工程实例研究,取得了以下研究成果:①对重庆山区中梁山、万盛、荣昌、天府、綦江等矿区代表性的矸石山进行详细踏勘,对矸石山的堆放形态及其区域的工程水文地质情况进行调查;针对重庆山区矸石山,测试矸石散体的颗粒级配、含水量、天然密度、干密度等参数、并在此基础上,通过现场剪切、压缩、渗透试验等研究矸石散体的力学特征。

And internal bond strength, modulus of rupture, modulus of elasticity, formaldehyde emission content, thickness expansion rate of water absorbing and density of MDF are analysed. The minor core protein s C-encoding gene of Muscovy duck reovirus was cloned into theprokaryotic expression vector pET32a. The recombinant plasmid pET32a-s C was amplified andextracted after being transformed into E.coli DH5a competent cells. Restriction analysis withEcoRⅠand SacⅠand sequences analysis indicated that the recombinant plasmid was inserted withcorrect open reading frame. The fusion protein about 50 ku was produced after induction with 0.15mmol/L IPTG of E.coli competent cells transformed with pET32a-sC. The SDS-PAGE andWestern-bloting test indicated that the fusion protein reacted with the convalescents sera of duckinfected with Muscovy duck reovirus. The indirect ELISA method was developed by using thepurified fusion s C protein as coating antigen. The optimal concentration of s C was 5μg/ml, thedilution of serum sample was 1:40; The results showed that preparation of an ELISA by using sCas coating antigen in detecting 50 field duck sera in comparison with the AGIP were more sensitiveand specific than agar gel immuno-diffusion AGIP test. The results suggest that presence ofantibody against viral protein sC in duck may be a good indicator by the sC-ELISA for detectionof duck infection with reovirus.
同时,本研究将编码外壳蛋白σC的基因克隆于原核表达载体pET32a上,经过EcoRⅠ和SacⅠ双酶切鉴定和序列分析后,得到阳性重组质粒pET32a-σC;将阳性重组质粒pET32a-σC转化到大肠杆菌BL-21感受态细胞中进行诱导表达,经SDS-PAGE和Western-blbtting检测分析,融合表达的蛋白能够与番鸭呼肠孤病毒感染的康复鸭血清发生特异性反应;将融合表达的蛋白纯化后作为包被抗原,建立了检测鸭血清中呼肠孤病毒抗体的间接酶联免疫吸附试验检测方法,此方法中抗原的最佳包被浓度为5μg/ml、标准阳性血清的最适稀释倍数为1:40倍,用此方法对50份鸭血清样品进行检测,并与琼脂糖凝胶扩散试验检测抗体的法相比较,证明此ELISA方法具有良好的特异性和敏感性,本研究为今后鸭呼肠孤病毒诊断试剂盒的研制奠定了基础。

Reliable testing basis is offered to analyze further failure and stability of the noncircular cave surrounding rock under high confining pressure and high hydraulic pressure.

According to the standard and request of mechanical structures for electronic equipment tests, the finite element model of the electronic equipment cabinet was built by using the software of finite element analysis-ANSYS and simulated the tests of random vibrations by loading and calculating. The 3-D isopleths graph of deformation and stress of the cabinet was obtained. It could confirm the maximum of deformation and stress of the cabinet and probability of occurrence. Through more analysis of the frequency response curve reflected directly and clearly the deformation of the cabinet within the particular seismic frequency.

The results showed compared with the native state the shear strength of rockmass under the saturation state decreased by 11.17%, cohesion force decreased by 16.09%, internal frictional angle decreased by 10.02%.(3) 3D-σFEM numerical simulation software is used to simulate the slope stability on the native state and saturation state. Compared with the nature state the displacement of the discontinuity and stress concentration in the saturation state is largened clearly, the results showed slope stability under the rainfall is weakened.
