Our three groups in SCBB will conduct one project in this grant (three projects in this grant total), which is "Screen mutants interfering the development or regeneration of endodermal organs and study their functional mechanisms". In this project, stem cell and zebrafish will be used as models to elucidate the mechanisms of the development of endodermal organs.

我院的三个课题组负责此项目的课题三《正向遗传突变筛选具有内胚层器官发育或再生缺陷的突变体及致变基因的功能机制研究》，将采用干细胞和斑马鱼模型，研究内胚层器官的机制，特别是分子调节网络的整体性和时空性，为防治肝脏和胰腺的重大疾病提供坚实的理论基础。

METHODS AK was purified by DEAE ion-exchange cellulose(DEAE-52) and sepharose Cl-4B .The component of the saccharide was determined by GC and TLC.Its MW was measure d by gel filtration.The form of glucosidic bond was detrermined by IR.The conten t of the saccharide was measured by colorimetry.

采用DEAE纤维素(DEAE-52)柱层析和凝胶柱层析分离纯化草乌多糖，气相色谱法和薄层层析法测定其多糖组成，凝胶过滤法测定其相对分子质量，红外光谱测定其苷键类型，比色法测定其糖含量。

Through PCR and sequencing, a fragement sequence with lamin gene feature was obtained. It was of of transcriptional activity. In addition, we also cloned and identified giardial DNA topoisomerase gene.Our phylogenetic analysis showed that although Giardia diverged early but not first, thus, it is not as primitive as thought before.

然后进行了核纤层蛋白基因的扩增和鉴定，获得了一个具有明显核纤层蛋白基因特征的基因序列片段，并证明它是表达的且不具内含子；此外还克隆鉴定了作为核骨架重要成分的DNA拓扑异构酶II的基因；在此基础上结合已报导的不同进化地位生物上的数据进行了分子系统学分析，结果显示贾第虫虽然分支较早，但不象过去所认为的那么原始。

By tailoring the scale between hydrophilic region and lipophilic region, smectic, columnar and cubic mesophases were observed in compounds I-F and II-F. By changing the length of the lateral chains, a series of new columnar phases and a non-conventional semctic phase were obtained in III-F.
