分光光度法

This paper is made up of 6 chapters; chapter 1 summarizes the interaction of low molecular weight compounds with DNA; chapter 2 to 6 focus on the electrochemical studies of some organic dyes such as pyronine B, phenosafranine, brilliant cresyl blue, crystal violet, neutral red etc. and their interaction with DNA, respectively. Different electrochemical methods such as linear sweep voltammetry, cyclic voltammetry and UV-Vis spectrophotometric method are applied to the investigation.

论文由6章组成，第1章综述了小分子和DNA的相互作用；第2至6章分别筛选了吡啰红B、酚藏花红、灿烂甲酚蓝、结晶紫和中性红等小分子染料，以汞电极如静汞电极、滴汞电极作为工作电极，采用循环伏安法、线扫极谱法等电化学手段，结合紫外-可见分光光度法，研究了它们的电化学行为以及和DNA的相互作用。

In order to comprehensively develop and utilize C. cathayensis exocarp and provide reference for protection of ecological environment at growing districts of C. cathayensis, inorganic constituents in C. cathayensis exocarp were analyzed by atomic absorption spectrometry. The results showed that content of K element in C. cathayensis exocarp was high, and the results of wet digestive and dry digestive methods were same.

为了综合开发利用山核桃外果皮，为山核桃产区生态环境的保护提供参考，经原子吸收分光光度法分析得知：山核桃外果皮的无机成分中含有较高的钾元素，山核桃外果皮可以作为制备工业钾盐的原料，湿消化法和干消化法处理外果皮所得的结果基本一致。

While erthrocyte, hemoglobin and Hela cell were under oxidative damage induced by the hydrogen peroxide (H2O2) or tert-butyl hydroperoxide on different concentrations of dihydromyricetin, hemolysis rate of erythrocyte and methemoglobin converted from hemoglobin was estimated by ultraviolet-visible absorption spectroscopy. Whereas mmalondialdehyde from erythrocyte was measured by thiobarbituric acid colorimetry, the cellular viability of Hela cell was examined using MTT 3-(4, 5-dimethylthiazol-2-yl-2, 5-diphenyl tetrazolium bromide assay.

The contents of potassium, sodium, calcium, magnesium, iron, chromium, nickel, manganese, copper and zinc in 11 kinds of traditional Chinese medicines, such as Albizia flower, milkvetch root, grassleaf sweetflag rhizome, thinleaf milkwort root, hawthorn fruit, ginkgo leaf, ginseng, Iongan pulp, cattail pollen, spine date seed and sappan wood were determined by atomic absorption spectrophotometry.

用原子吸收分光光度法检测合欢花、黄芪、石菖蒲、远志、山楂、银杏叶、人参、元肉、蒲黄、酸枣仁、苏木11种中药中钾、钠、钙、镁、铁、铬、镍、锰、铜、锌元素的含量，结合锌/铜比值、镁/钙比值，以数据极差法进行统计指标的标准化处理，以数量积法算出衡量被分类对象之间的相关系数，确定论域上的模糊矩阵。

Commonly used method is: EDTA titration method, atomic absorption spectrometry and potassium permanganate titration.

Methods］ MTT assay was employed to test the lethal concentration 50 (IC50) of sensitive germline (SK-OV-3) and ovarian cancer cell subline induced by ADR (SK-OV-3/adr) treated by 4 drugs, resistance index and reverse multiple were calculated. The changes of drug content were observed by fluorescence microscope, the intracellular accumulation of ADR was evaluated by fluorospectrophotometry, the changes of mdr 1/P-gp resistance were observed respectively by RT-PCR and Western blot.
