Methods: During the April 2003 to December 2004, from the patients with different position colorectal cancer beyond Dukes C stage who were performed laparoscopic radical operation random draw 20 patients as experiment group, and from patients with different position colorectal cancer beyond Dukes C patients who were performed open radical operation 20 patients took randomly as contrast group. We draw thepatients blood about 5 ml from antecubital vein at the time before and after the operation about 30 minutes respectively, preserved the blood in deep hypothermia circumstance,cytrokeratin-20(CK-20) as the micrometastasis maker, using the RT-PCR technique,G3PDH as the intra-contrast gene, quantitatively detect the amounts of CK-20 in the blood with the gelatum imagery system and the computer analyzing software on the gelatum imagery system ,in order to compare the influence of the laparoscopic and the open operation on the micrometastasis.

在我院2003年4月—2004年12月间不同部位的临床分期在Dukes C期之内的接受了腹腔镜结直肠癌根治术结直肠癌患者中随机抽取20例作为试验组，另外在同期的接受开腹手术患者中随机抽取20例作为对照组，分别抽取术前30min和术后30min的肘前静脉血5ml，集中低温保存，以细胞角蛋白(cytekeratin-20)CK-20为微转移的标志物，应用RT-PCR技术，以G3PDH为内对照基因，应用凝胶成像系统及其计算机凝胶成像系统分析软件来定量检测手术前、后患者外周静脉血中CK-20含量，比较腹腔镜和开腹的手术方式对肿瘤微转移的影响。

RESULTS: A well-resolved and reproducible 2-DE pattern of PBMC was obtained from patients with HBV associated HCC or LC. For HCC, the mean number of protein spots was 1186±43, with an average matching rate of 91.6%, and for LC, the mean number of protein spots was 1013±41, with an average matching rate of 90.2%. Twenty-seven differential proteins were tested by MALDI-TOFMS, and 16 were identified. Of the 16 proteins, 13 were up-regulated in patients with HCC, including heat-shock protein 27, member RAS oncogene family (RAB14), actin, alpha 1 antitrypsin, RNAbinding protein regulatory subunit and so on. By contrast, the levels of proteins such as PDX6, HSPA8 and MnSOD were significantly reduced in HCC patients.

结果：得到了分辨率较高、重复性较好的HBV相关性HCC及LC患者PBMC双向凝胶电泳图谱，HBV相关性HCC及LC患者PBMC凝胶的平均蛋白质点数分别为1186±43及1013±41；组内的平均匹配率分别为91.6%，90.20%对27个差异蛋白质点进行了质谱分析，鉴定出16个蛋白质，在HBV相关性HCC患者PBMC中表达明显增强的有热休克蛋白质27、ras癌基因家族(RAB14)、肌动蛋白、α1-抗胰蛋白及RNA结合蛋白调节亚基等13个蛋白点；而PDX6、HSPA8及锰超氧化物歧化酶在HCC患者PBMC中低表达。

Isolations of the s.aureus soluble peptidoglycan and quality control:(1) the s.aureus soluble peptidoglycan was released by short dose benzylpenicillin sodium and purified by sephadex G-100.(2) The quality control in the process of isolation: the soluble peptidoglycan was detected by SLP reagent kit; the LPS was detected by kinetic turbidimetric limulus test;and the protein was detected by improved LOWRY reagent kit.

金黄色葡萄球菌可溶性肽聚糖的分离提取及其质量控制(1)采用青霉素钠诱导法和葡聚糖G-100(Sephadex G-100)凝胶柱分离提取金黄色葡萄球菌可溶性肽聚糖；(2)金黄色葡萄球菌可溶性肽聚糖提取过程中的质量控制：采用SLP试剂盒对提取物定性检测，应用动态浊度法鲎试验测定提取物中内毒素含量，采用改良LOWRY试剂检测提取物中蛋白的含量；(3)采用青霉素诱导和Sephadex G-100凝胶柱分离提取金黄色葡萄球菌可溶性肽聚糖的条件：青霉素使用量、加入青霉素后孵育时间和菌液浓度等优化选择。
