Result The detection limit of the two systems were 0.1fg, equal to 2.31×10^4 copies, which was 10^2-10^3 fold higher than conventional PCR. By the constructed detection systems, the TCK and Tilletia cariesTul could be distinguished. The teliospore and mycelium of TCK in the infected wheat plant tissue also could be identified accurately and rapidly.

建立的两套定量PCR检测体系的检测下限相当，可达到0.1fg，对应的拷贝数为2.31×10^4个，检测灵敏度比常规PCR高2~3个数量级，均可成功鉴别出TCK与小麦网腥黑穗病菌Tilletia caries (DC Tul， TCT，并可快速准确检测小麦矮腥黑穗菌冬孢子和检测罹病小麦植株体内的侵染菌丝体。

In this reseach, we designed specific primer pairs based on the species-specific DNA fragment of TCK (1322 bp) from telomere associate gene of TCK and successfully established the routine PCR and real-time PCR detection system for teliospore or mycelium of TCK, which will facilitate accurate detection and identification of TCK teliospore on seeds, soil, flower and feeds and infected mycelium inside of wheat tissue for development scientific strateges of desease risk assessment and management.

本研究以筛选到的1322bpTCK独有差异基因设计TCK特异性引物对，建立TCK常规和定量荧光PCR技术体系，并研制出快速检测试剂盒。其研究成果将有助于对种子、土壤、原粮加工副产品、饲料和废弃物上病菌冬孢子流向和存活力进行动态监测，为制定科学的病害风险管理策略和监控方案提供技术支撑。
